小鼠血清脂联素水平与脂肪性肝病

摘　　要：目的研究高脂及高脂加酒精饮食诱导的实验性小鼠在脂肪性肝病形成不同阶段血清脂联素水平的变化情况。方法将小鼠分为3组：对照组、高脂饮食模型组、高脂加酒精饮食模型组。检测不同阶段小鼠血清脂联素、甘油三脂（Triglyceride，TG）、丙氨酸氨基转移酶（Alanine aminotransferase，ALT）、胰岛素敏感指数（Insulin sensitive index，ISI），并观察肝脏组织病理学变化。结果高脂饮食5周及7周小鼠血清脂联素分别为10．52±1．28μg／ml、8．85±1．42μg/ml，均明显低于对照组12．36±0．80μg/ml（P〈0．01）。高脂加酒精饮食5周及7周小鼠血清脂联素分别为9．04±1．08μg/ml、8．16±1．85μg/ml均明显低于对照组12．05±1．53μg/ml（P〈0．01）。血清脂联素与TG、ALT、脂肪性肝病炎症活动度负相关，与ISI正相关（r值分别为-0．830，-0．801，-0．728，0．847）.结论随着脂肪性肝病的形成与发展，脂联素水平逐渐降低。脂联素将可能成为衡量脂肪性肝病严重程度及临床预后判断的一个血清学指标。AIM To study the change of serum adiponectin at different phase of mice fatty liver model induced by high fat diet and high fat ethanol diet. METHODS Fifty- two ICR mice were divided into three groups namely control group, high fat diet model group,high fat ethanol diet model group,to test the level of serum adiponectin,triglyceride （TG） ,alanine aminotransferase（ALT） ,insulin sensitive index （ISI）, and pathologic changes in liver at different phase of mice fatty liver model induced by high fat diet and high fat ethanol diet. RESULTS The serum adiponectin was significantly reduced in the model groups compared with the control group at 5 weeks[ （10.52 ± 1.28）μg/ml vs （12.36 ± 0. 80）μg/ml, （ P 〈 0. 01） ; （8.85 ± 1.42）μg/ml vs （12.36 ± 0. 80）μg/ml, （ P 〈 0. 01 ）] and 7 weeks（9.04 ± 1.08）μg/ml vs（12.05 ± 1.53）μg/ml, （ P 〈 0. 01） ; （8.16 ± 1.85）μg/ml vs. （ 12.05 ± 1.53） μg/ml, （ P 〈 0. 01）. There was a negative correlation between the level of TG,ALT,HAI and the serum adiponectin,on the other hand,the adiponectin level positively correlated with ISI. CONCLUSION With the development of fatty liver disease, the serum adiponectin decrease. To some extent, it can reflect fatty liver degree in mice.
