摘  要:目的:通过对重组人干扰素-γ(rhIFN-γ)供试品与rhIFN-γ参考品和重组人干扰素-α2a(rhIFN-α2a)参考品圆二色谱图形比较,进而验证他们的结构是否一致。方法:分别独立处理rhIFN-γ供试品和rhIFN-γ参考品3次,每次分别在190~240nm波段(远紫外区)和250~320nm波段(近紫外区)用圆二色谱测量,对得到的6组远紫外区数据和6组近紫外区数据,分波段进行相关性分析,然后对相关系数做统计分析;再用同样方法,对rhIFN-γ供试品和rhIFN-α2a参考品圆二色谱图形对比分析。结果:rhIFN-γ供试品和rhIFN-γ参考品在远紫外区和近紫外区圆二色谱图形没有差别;但rhIFN-γ供试品和rhIFN-α2a参考品在2个波段圆二色谱图形都有明显差别。结论:在远紫外区rhIFN-γ供试品和rhIFN-γ参考品圆二色谱图形的一致,说明两者二级结构比例相同,在近紫外区圆二色谱图形的一致,说明两者侧链生色团的排布相同,从而说明两者结构相同;而rhIFN-γ供试品和rhIFN-α2a参考品在远紫外区和近紫外区两者圆二色图形均不一致,说明二者在结构上不同。Objective:To identify the structure coherence between the sample of recombinant human interferon gamma ( rhIFN-γ) and the rhIFN-γ reference and the structure differences between the rhIFN-γ sample and the reference of recombinant human interferon alpha 2a ( rhIFN -α2 spectra graphs. Method: The rhIFN-γ sample and the rhIFN -α) through comparing circular dichroism (CD) reference substance were independently pretreated for three times respectively. The rhIFN-γ sample and the rhIFN-γ reference were respectively detected under 190 -240 nm (far UV) and under 250 -320 nm (near UV) by CD at a time. Six serieses of data in the far UV group and in near UV group were obtained respectively. The correlation coefficients were calculated for each wave band and the differences were estimated by statistical test. Moreover, the CD graphs of the rhIFN-γ sample and the rhIFN -α2a reference substance were compared through the same method. Result:There were no differences of CD graphs found between the rhIFN-γ sample and the rhIFN -γ reference substance in the far UV and in the near UV. But there were differences between the rhIFN-γ sample and the rhIFN - α2a reference substance. Conclusion:The graph coherence between the rhIFN -γ sample and the rhIFN -γ reference substance in the far UV hinted that the second structure proportion is same and the graph coherence in the near UV hinted that the chromophores distribution is same. According to the above, there are the same protein stuctures between them. But the graph differences between the rhIFN-γ sample and the rhIFN - α2a reference substance hint that their protein structures are different.
