肾衰养真胶囊对5/6肾切慢性肾衰营养不良大鼠瘦素及其受体mRNA表达的影响

摘　　要：目的:观察5/6肾切除大鼠营养不良状态;以及肾衰养真胶囊对慢性肾衰竭(CRF)营养不良大鼠瘦素(Lep-tin)及其受体mRNA表达的影响。方法:5/6肾切除制作CRF大鼠模型,同时予4%酪蛋白饮食,检测尿素氮、肌酐、白蛋白等生化指标和体重观察其营养不良发生时间,符合条件的随机分为模型组、肾衰养真胶囊治疗组和开同治疗组;另设正常对照组。灌胃6周后应用放射免疫法检测血浆Leptin水平和RT-PCR法检测下丘脑瘦素受体(OB-Rb)表达。结果:CRF大鼠在术后10周末出现营养不良,肾衰养真胶囊可减低增加血浆白蛋白、血红蛋白;肾衰养真胶囊可下调OB-Rb表达,降低Leptin水平。结论:肾衰养真胶囊可能通过调节下丘脑OB-Rb活性,从而进一步改善CRF大鼠营养不良。Objective ：To investigate the malnutrition situation in rats by 5/6 nephrectomy and effect of Shenshuai Yangzhen capsule,a preparation of traditional Chinese medicine, on leptin and OB - RbmRNA in malnutrition rats with chronic renal. Methods ：SD rats were received 5/6 nephrectomy for preparation of CRF models, and fed 4% casein at the same time. The blood parameters, like BUN, SCr, Alb and weight in CRF rats were determined, observed when malnutrition began. Those in condition were randomized into CRF group, kaitong group, Shenshuai Yangzhen group, and normal control group. After 6 -week treatment as indicated, subthalamencephalon OB -Rb messenger RNA （OB -RbmRNA） level were measured by RT-PCR in rats with surgically induced renal failure （two- stage subetotal nephrectomy）. The blood leptin in CRF rats were determined by radio immunoassay. Results： Malnutrition situation in CRF rats began at the end of 10 -week. Compared with those in the CRF group, the plasma leptin concentrations and OB - Rb gene expression in Shenshuai Yangzhen group were significantly Lower with substantially elevatel plasma Alb and Hb. Conlusion： Shenshuai Yangzhen capsule can improve malnutrition in rats with renal insufficiency possibly by Depressing OB -Rb gene expression.
