摘 要:目的:探讨姜黄素对体外培养人卵巢癌HO-8910细胞增殖和凋亡的影响。方法:选用人卵巢癌细胞株HO-8910进行体外培养至对数生长期,以0-80μmol/L姜黄素(5μmol/L,10μmol/L,20μmol/L、40μmol/L、80μmol/L)分别处理HO-8910细胞24、48、72h,四甲基偶氮唑蓝(MTT)实验测定细胞的生长抑制率;流式细胞仪检测不同浓度药物对细胞凋亡率的影响;免疫组化法检测PCNA在细胞中的表达,评价细胞的增殖状态。结果:姜黄素对HO-8910细胞增殖抑制率随药物浓度增加及作用时间延长有升高趋势,呈时间剂量依赖性。姜黄素作用24h随浓度的升高PCNA阳性表达逐渐降低。流式细胞仪分析证实随着浓度的升高细胞凋亡率和膜受损率均提高。结论:姜黄素对人卵巢癌细胞株HO-8910的生长有显著的抑制作用,并能诱导其凋亡。Objective: To investigate the effects of curcumin on proliferation and apoptosis of human ovarian cancer cell line HO-8910 in vitro. Methods: The human ovarian cancer cell line HO-8910 was cultured to logarithm growing period and treated with curcumin of different concentration(5μmol/L, 10μmol/L,20μmol/L,40μmol/L,80μmol/L). The inhibition rates of ceils were detected through MTT assay; the apoptosis rates of cells was tested by flow cytometry; the expression of PCNA in HO-8910 cell was observed by SABC imunohistochemistry. Results. Curcumin inhibited the proliferation of HO-8910 calls in a dose-and time-dependent manner. The tumor cell apopotic rate in curcumin group was significantly higher than those in control group. The expression of PCNA was increased in a dose dependent manner. Conclusion:The curcunm not only inhibits proliferation, but also induces apoptosis of human ovarian cancer cell line HO-8910.
