摘  要:目的探讨 K-Cl 协同转运子1(KCC1)在宫颈癌组织中的表达及在宫颈癌发生、发展中的作用。方法采用半定量逆转录-聚合酶链反应(RT-PCR)、Western 印迹及免疫荧光染色方法测定40例宫颈癌、10例宫颈上皮内瘤样病变(CIN)和10例慢性宫颈炎组织中 KCC1mRNA 及蛋白的表达及定位情况。结果宫颈癌组织中 KCC1mRNA 和蛋白的表达(灰度比值分别为1.29±0.17,1.18±0.27)均明显高于慢性宫颈炎(0.59±0.27,0.33±0.18)、CIN 组织(0.50±0.28,0.27±0.15),差异有统计学意义(均 P<0.05)。宫颈低、中分化癌组织中 KCC1mRNA 及蛋白的表达(灰度比值分别为1.39±0.12,1.37±0.15)明显高于高分化组(1.16±0.14,0.96±0.19),差异有统计学意义(P<0.05);KCC1mRNA 及蛋白在晚期宫颈癌组织中的表达与早期组差异无统计学意义,KCC1蛋白在宫颈癌及对照组织中的分布定位在细胞膜上。结论 KCC1基因可能在宫颈癌的发生发展中起重要的作用。Objective To investigate the expression of K-Cl cotransport 1 ( KCCl ) in cervical cancer tissues and to investigate its role in the onset and progression of cervical cancer. Methods Specimens of uterus were obtained from 60 patients aged 45.89 ( 25 - 73 ), 40 with cervical cancer, 10 with cervical intraepithelial neoplasia (CIN), and 10 with chronic cervicitis. Semi-quantitative RT-PCR was used to detect the mRNA expression. Western blotting was used to detect the protein expression of KCCl. assay was used to detect the location of KCCl protein. Results The mRNA expression and protein expression of KCCl were both significantly higher in the cervical cancer tissue than those in the CIN and chronic cervicitis tissues ( all P 〈 0. 05 ). The levels of KCCl in the lowly differentiated cancer tissues (at grades G2 and G3 ) were significantly higher than those in the highly differentiated cancer tissues (at grade G1, P 〈 0. 05 ). Western blotting revealed that the protein expression level of KCCl was significantly higher in the cervical cancer tissues than in the CIN and chronic cervicitis tissues (both P 〈 0. 05) and the protein expression levels of KCCl in the cancer tissues at G"2 and G3 grades were significantly higher than that in the cancer tissues at grade G1 ( both P 〈 0.05 ). There were no significant differences in mRNA and protein expression between the early and terminal cervical cancer tissues. There were no significant differences in the mRNA and protein expression Of KCCl between the cervical cancer cases with or without lymph node metastasis. Immunofluorescence assay showed that KCCl was located in the cellular membrane in all patients and the KCCl expression level there was significantly higher in the cervical cancer tissues than in the tissues of CIN and chronic cervititis. Conclusion The expression of KCCl is upregulated in cervical cancer and it may play an important role in the onset and progression of cervical cancer.
