结直肠癌组织中葡糖醛酸基转移酶2B表达水平的研究

摘　　要：目的检测葡糖醛酸转移酶2B家族(UDP-Glucuronosyltrans-ferase 2B,UGT2B)在结直肠癌组织中的表达,探讨其在结直肠肠癌变中的意义。方法病例组32例结直肠癌组织、癌旁组织及对照组14例正常人群黏膜,用逆转录多聚酶链式反应(RT-PCR)方法检测葡糖醛酸转移酶2B家族(UGT2B)mRNA表达。Western blot方法检测UGT2B7蛋白在结直肠癌组织、癌旁组织及正常结肠黏膜内的表达。结果UGT2B在结肠黏膜内呈多态性表达;与对照组相比,结肠癌组织、癌旁组织UGT2B4mRNA水平无明显改变,UGT2B7、UGT2B15mRNA表达水平降低(P<0.05);结直肠癌组织、癌旁组织UGT2B7蛋白蛋白灰度值比值(分别为:0.186±0.109和0.409±0.171)低于正常结肠黏膜0.684±0.117(P<0.05)。结论与正常结肠黏膜比较,结直肠癌组织、癌旁组织UGT2BmRNA和蛋白表达水平降低。UGT2B同工酶表达降低或缺失可能是结直肠癌变的机制之一。建议具有低活力UGT酶表达的个体应远离接触致癌物的工作,并戒烟,对于防止这些高危人群结直肠癌的发生,有重要的现实意义。Objective To detect the expression of UDP-glucuronosyltransferase （UGT） 2B subfamily in colorectal cancer, and to investigate its significance in colorectal tissue carcinogenesis. Methods Reverse transcription and polymerase chain reaction （RT-PCR） were used to determine the mRNA levels of four UGT isoforms in 32 specimens of colorectal cancer, 14 specimens of normal colorectal tissues. Western blot analysis was performed for detection of colonic UGT2B7 protein in situ. Results Polymorphic regulation ofUGT2B isoforms expressed in human colorectal tissue; compared to control group, UGT21M mRNA level of the 32 samples of colorectal cancer and its surrounding healthy tissue was not significantly changed; UGT2B7 mRNA level was significantly down-regulated in colorectal cancer and its surrounding healthy tissue; compared to control group, UGT2B7 protein level was significantly down-regulated in colorectal cancer and its surrounding healthy tissue. Conclusion UGT2BmRNA expression and protein expression level were decreased in colorectal cancer tissues and its surrounding healthy tissue. This implies that interindividual regulatory mechanisms may be the biological basis that determines interindividual variations in cancer risk. We advise these individuals who express low UGT enzyme activity should avoid working in the enviorment of interacting with poison, and stop smoking. It can make effect in decreasing colorectal cancer incidence, and have important clinical significance.
