高效液相色谱梯度洗脱法同时测定阿魏酸和天麻苷的含量

摘　　要：采用高效液相色谱梯度洗脱法同时测定阿魏酸和天麻苷的含量,以C18-ODS为色谱柱,甲醇-1%冰醋酸溶液为流动相进行梯度洗脱,阿魏酸和天麻苷的检测波长分别为323nm、270nm,柱温:25℃。结果表明阿魏酸在0.09-0.9μg范围内有良好的线性关系(r=0.9999),天麻苷在0.85-8.5μg范围内有良好的线性关系(r=0.9999),测得滴丸中阿魏酸含量3.72mg.g-1,天麻苷含量4.28 mg.g0-1。本实验在30m in一次进行测定复方天麻滴丸中两个标志性成分,简便、快速、结果可靠。The contents of ferulic acid and gastrodin were simultaneously determined by gradient elution of HPLC. Methanol - 1% glacial acetic acid was used as mobile phase, gradient elution was carried out with C18 -ODS chromatogram column. The detection wavelength of ferulic acid and gastrodin was 323nm and 270nm respectively and the column temperature was 25℃. The results indicated that the ferulic acid of calibration curves were linear at the ranges of 0.09 -0.9μg （r =0.9999）, the gastrodin of calibration curves were linear at the ranges of 0.85 -8.5μg （r =0.9999）. In addition, the content of ferulic acid and gastrodin in soft capsule was 3.72 mg · g^ -1 and 4.28 mg · g^-1 respectively. This method could determine two typical components in soft capsule in 30 minutes in single test, and it was simple, fast and reliable.
