微米中药胰腺康诱导人胰腺癌SW1990细胞凋亡的研究

摘　　要：目的:观察不同浓度的微米中药胰腺康对胰腺癌SW1990细胞作用的影响。方法:将微米中药胰腺康以不同浓度和时间作用于胰腺癌SW1990细胞,采用MTT比色法观察细胞毒性;应用HE染色法观察凋亡细胞的形态;采用免疫组化方法检测caspase-3蛋白的表达。结果:微米中药胰腺康以剂量依赖和时间依赖的方式抑制SW1990细胞的生长。HE染色观察到凋亡细胞的形态学改变。caspase-3蛋白高表达。结论:微米中药胰腺康与其提取液相比,能更有效地抑制SW1990细胞增殖,其抗瘤作用与诱导细胞凋亡有关。为临床治疗胰腺癌提供理论依据。Objective： To investigate the effects of different concentrations of micrometer Chinese traditional drug of Yixiankang on human pancreas carcinoma cell line SW1990. Method： The pancreas carcinoma cell line SW1990 was treated with micrometer Chinese traditional drug of Yixiankang at its different concentrations for different duration, and cytotoxicity was evaluated by methabenzthiazuron （MTT）method,and apoptosis-related alterations in morphology were dertermined with haematine-eosin （HE）staining. The expression of caspase-3 was detected by immunohistochemistry. Results：Micrometer Chinese traditional drug of Yixiankang inhibited the growth of pancreas carcinoma ceils in a dose and time dependent manner. Haematinc-eosin （HE） staining showed that the cells treated with micrometer Chinese traditional drug of Yixiankang exhibited characteristics of apoptosis. The expression rate of caspase-3 was higher. Conclusion ：Yixiankang compound can inhibit the proliferation of pancreas carcinoma cells,and the micrometer Chinese traditional drug is more efficient than the extraction of Yixiankang. The anti-tumor effect is associated with induction of cancer apoptosis.
