摘  要:目的:探讨环氧化酶-2(COX-2)选择性抑制剂塞来昔布对人子宫内膜腺癌细胞系HEC-1B侵袭力的影响及其相关机制.方法:用噻唑蓝(MTT)法检测不同浓度塞来昔布作用HEC-1B细胞不同时间后细胞增殖的变化;不同浓度塞来昔布作用HEC-1B细胞24h后,Transwell小室法测定细胞的侵袭力,RT-PCR法测定细胞中COX-2 mRNA的表达.结果:MTT检测显示塞来昔布对HEC-1B细胞的增殖抑制作用呈时间依赖性和浓度依赖性;不同浓度塞来昔布作用HEC-1B细胞24h,Transwell侵袭实验结果表明,在50~200μmol/L塞来昔布组随着浓度的增加细胞穿膜数逐渐减少(P<0.05),RT-PCR结果显示,50~200μmol/L塞来昔布组随着药物浓度的增加COX-2 mRNA的表达逐渐减弱(P<0.05).结论:塞来昔布能抑制HEC-1B细胞增殖,降低细胞的侵袭力,其机制可能与下调细胞中COX-2 mRNA的表达相关.Objective :To investigate the influence of cyclooxygenase-2 selective inhibitor celecoxib on the invasion of human endometrium adenocarcinoma HEC-1B cell line and the potential mechanism. Methods:The HEC-1B cell proliferation was examined by MTF assay. After being treated with different concentrations of celecoxib for 24hours, the cells invasion was detected by transwell chamber,the expression of COX-2 mRNA was analyzed by RT-PCR. Results:MTF results indicated that celecoxib could inhibit the proliferation of HEC-1B cells in a time- and dose-dependent manner. Treated with different concentrations of celecoxib for 24hours, transwell chamber assay indicated that the invasion of cells gradually decreased with the increase of celecoxib dose (50 - 200μmol/L group, P 〈 0. 05 ). RT-PCR results indicated that the expression of COX-2 mRNA gradually decreased with the increase of celecoxib dose( P 〈 0.05 ). Conclusion:Celecoxib can inhibit the proliferation of HEC-1B cells and depress invasion of the cells. The mechanism may be associated with down-regulation of COX-2 mRNA expression in HEC-1B cells.
