喉癌中SSTR2和SSTR5 mRNA的表达及意义

摘　　要：目的研究生长抑素受体(somatostatin receptor,SSTR)亚型SSTR2和SSTR5在喉癌组织及癌旁正常组织中的表达,以及与临床病理特征的关系。方法应用反逆转录-聚合酶链反应(RT-PCR)方法,检测46例喉癌组织与30例癌旁正常组织中SSTR2和SSTR5的表达,分析其与临床病理特征的关系。结果在46例喉癌中SSTR2和SSTR5的表达率分别为58.70%和82.61%;在癌旁正常组织中的表达率分别为83.33%和90.00%。喉癌中SSTR2的表达低于癌旁正常组织(P<0.05);SSTR5的表达与癌旁正常组织无统计学意义(P>0.05)。SSTR2和SSTR5的表达与喉癌的临床分期、病理分化程度明显相关(P<0.001)。结论大多数喉癌肿瘤组织中有一种以上生长抑素受体亚型基因的表达,其表达随喉癌恶性程度呈下降趋势。Objective To study the expressions of somatostatin receptor subtype 2 （SSTR2） and 5 （SSTR5） in human laryngeal carcinoma and its adjacent normal tissue and to explore the relationship between the expression and clinicopathological characteristic. Methods Reverse transcription-polymerase chain reaction （RT-PCR） was used for defecting the mRNA levels of SSTR2 and SSTR5 in 46 specimens from laryngeal carcinoma and 30 specimens of adjacent tissue The relationship between the expression and clinicopathological characteristic was also analyzed. Results The positive expression rates of SSTR2 and SSTR5 in carcinomatou tissue were 58. 70% and 82. 61% respectively, while those in the adjacent tissue were 83. 33% and 90%. There was no significant difference in the positive rates of SSTR5 between carcinomatous tissue and adjacent tissue （P 〉0. 05）, but the positive rate of SSTR2 in carcinomatous tissue was significantly lower than that in adjacent tissue （ P〈0. 05）. The significant correlations were found between SSTRs expressions and pathological stages （ P〈0. 01） and differentiation degrees （ P〈0. 01） of the tumor. Conclusion More than one SSTR subtype genes are expressed in laryngeal carcinoma and the expression declines with the increase of malignant degree of the tumor.
