摘 要:目的评价坎地沙坦酯对实验性有色家兔脉络膜新生血管(CNV)血管内皮生长因子(VEGF)表达的抑制作用。方法24只健康有色家兔,实验眼通过氩激光破坏性光凝诱导脉络膜新生血管模型,然后随机分成两组,给予不同处置。A组为对照组;B组为坎地沙坦酯组。分别于处置后24h,7、14、28d散瞳并麻醉后行光学相干断层扫描(OCT)、荧光素眼底血管造影(FFA)检查后,过量麻醉处死动物取眼球后节行组织病理学检查,并行血管内皮生长因子(VEGF)免疫组织化学分析。结果两组实验眼随处置时间的不同,各项检查有不同的变化,7d时表达最高,与24h,14d、28d比较差异有统计学意义(P<0.05);且治疗组表达低于对照组,差异有统计学意义(P<0.05)。结论坎地沙坦酯抑制实验性CNV中VEGF的表达。Objective Age-related macular degeneration (AMD) is the most common cause of legal blindness among the urbanized world. Choroidal neovascularization (CNV) is the hallmark of AMD and a leading cause of visual loss after aged 55. The present study was to investigate the inhibitory effect of candesartan cilexetil, an angiotensin Ⅱ receptor blockades drug, on experimental CNV in pigmented rabbits. Methods Twenty-four pigmented rabbits were randomly divided into control group and candesartan cilexetil group. The retina of rabbits in candesartan cilexetil group was photocoagulated by argon laser to induce the CNV and 5 mg/( kg · day) candesartan cilexetil was administrated orally for 1 day, 7, 14 and 28 days, and the only photocoagulation was used in normal control eyes. The effect of candesartan cilexetil was evaluated by the optical coherence tomography (OCT) and fluorescein fundus angiography ( FFA ) in various times above mentioned. Immunohistochemistry were performed for retinal morphological examination. Computer image analysis was used to analyze the expression of vascular endothelial growth factor(VEGF) in retina. Results Lesion rank of the fluorescence leakage was significant on day 28 after laser photocoagulation( P 〈 0.05 ). The mean thickness of the highly reflective layer by OCT in control group was significantly higher than candesartan cilexetil group. The mean gray scale values of VEGF in candesartan cilexetil group was significantly lower than that in control group. The mean gray scale values of VEGF at 7 days was significantly higher than that at 1 day, 14 and 28 days in two groups( P 〈 0.05). Conclusion Candesartan cilexetil can inhibit the development of laser-induced CNV and the expression of VEGF in rabbit.
