摘  要:本研究在研制出针对沙门菌鞭毛蛋白的属特异性单抗基础上,应用该单抗在免疫荧光、免疫胶体金试验、SDS-PAGE和免疫转印分析中,证实鞭毛蛋白携带该菌属共同抗原表位,并定位于F27片段上。通过分离,鉴定与表达鼠伤寒沙门氏菌Ⅰ相鞭毛蛋白基因fliC^i,又在基因水平揭示出该类表位的遗传背景。对该类抗原表位分布规律和抗原性的分析,阐明了它的属特异性和属内的广谱性、良好的免疫原性和多种表位的特性,这与分型H抗原表位明显不同。大量试验证实,该类表位可作为沙门氏菌属识别标志,为该菌快速检验和鉴定提供了新途径,为分类学研究提供了新证据。The multiple epitopes of generically-specific common antigen on Salmonella flagellins were defined by a panel of monoclonal antibodies in immunogold, Indirect immunofluorescent test, Western blotting and ELISAs. This result was further verified by detecting these epitopes on the expression product of flagellin gene fliCi in E. Coli Le—2a. The distribution of these epitopes was assessed by examining the binding patterns of each MAb to a set of 219 Salmonella strains and 892 isolates covering A through O—67 serogroups and 96 other enteric bacteria. The results showed that these common epitopes on Salmonella flagellin molecules could be used as a new recognized marker for the genus of Salmonella. These epitopes could be used for the identification of Salmonella isolates and rapid detection of Salmonella contamination in different specimens.
