摘  要:目的建立皮质发育不良(CD)大鼠模型,观察行为、脑电及大脑病理特征,探讨病理与致痫机制的关系。方法建立60Coγ-射线辐照诱导CD动物模型,视频监测;脑电;HE、Nissl、Timm's硫化银和细胞凋亡-Hoechst染色;电境观察。免疫组化采用SABC法,DAB显色。结果模型鼠少数自发性癫痫发作。额叶皮质及海马EEG示频繁自发的棘波、尖波、棘慢综合波等发放。病理主要表现为皮质和海马结构异常及细胞结构异常,双侧海马CA3区苔藓纤维发芽,海马CA1区结节中有致密浓染的凋亡细胞核。电境示CD鼠脑皮质及海马CA1区结节锥体神经元内线粒体异常、内质网扩张等。GABAARα1阳性细胞在Fr、HL、Par1、Par2、CA1、CA2、CA3区减少(P<0.05)。结论γ-射线诱导的皮质发育不良大鼠模型是模拟人类神经元异位、室周结节性异位的理想动物模型。Objective To establish^60 Co γ-ray extemal irradiated injury model of cortical dysplasia (CD) in rats,observe the changes of symptoms,EEG and pathology during the course of the model development,and discuss the meehamism of pathologic changes and epileprogenesis in this model. Methods Establish γ-ray-induced model of CD in rats. The symptom of the rats were recorded by a video camera. EEG signals were recorded by scalp electroencephalogram. The pathologic examination was performed, including HE, Nissl, Hoeehst and T imm's stanining to neuron,apoptosis and mossy fiber sprouting in different groups. Electron microscope was utilized to observe morphological changes in cortex and hippocampal formation. GABAARal-immunoreaetivity was studied in the cortex and hippocampal in rats and assessed the hippocampal mossy fiber sprouting after Timm's histochemical methods. Results The rats had seizure by chance. EEG showed frequent spontaneous sharp waves,or spikes from the frontal or the hippocampal neocortical structures. Neonatal γ-ray irradiated injury could result in CD in cerebral cortex and mossy fiber sprouting in hippoeampi CA3 were bilaterally. There were intensive hyperchromatic cell nu- cleus of apoptosis in the CA1 subfield. The eleetromieroseopie examination showed mitoehondria lesions,endoplas- mie reticulum expanding in neurons. There were changes of GABAARal subunit in cortical dyspiasia and hip- poeampi, Only the optical densities of GABAARal-immunoreaetive neurons located Fr, HL, Par 1, Par2,CAt, CA2, CA3area was significant between the F1 groups and the two control groups(P^0.05). Conclusion The γ-ray-induced CD model is an ideal animal model simulating human neuronal migration disorder.
