摘 要:目的观察TAGLN在胃癌中的表达及其与肿瘤细胞侵袭转移能力的关系。方法运用实时PCR、Western blotting及免疫组化方法,检测TAGLN在40例胃癌患者肿瘤组织和相应癌旁组织、7株胃癌细胞株及癌相关成纤维细胞(CAFs)中的表达;分析TAGLN的表达与胃癌临床病理学之间的关系。运用细胞侵袭和移动试剂盒检测高表达TAGLN的CAFs或沉默TAGLN表达后的CAFs对胃癌细胞株MKN45侵袭及移动能力的影响。结果在mRNA和蛋白表达水平方面,各胃癌细胞株间无显著性差异(P>0.05);与相应癌旁组织相比,胃癌组织TAGLN的表达明显上调(P<0.05);免疫组化染色显示,TAGLN蛋白表达于胃癌组织间质中的成纤维细胞、新生血管内皮细胞及肿瘤浸润的黏膜肌层中的肌细胞,且主要表达于细胞膜和细胞浆。淋巴结转移组胃癌组织TAGLN蛋白的表达为3.751±0.681,显著高于无淋巴结转移组的1.084±0.328(P<0.05)。细胞侵袭和移动试验结果显示,高表达TAGLN的CAFs能够促进胃癌细胞株MKN45的侵袭及移动能力;而经siRNA沉默TAGLN表达后的CAFs则明显降低胃癌细胞株MKN45的侵袭及移动能力。结论TAGLN基因在胃癌组织的基质中高表达,其表达上调对胃癌的侵袭和转移可能具有促进作用。Objective To observe the expression of TAGLN in gastric cancer and its relationship with tumor invasion and metastasis, Methods Real-time PCR, Western blotting and immunohistochemistry were applied to examine the expression of TAGLN in gastric cancer tissues and cancer-adjacent tissues (n = 40) , gastric cancer cell lines ( n = 7) and fibroblasts ( including CAFs) , respectively. The relationship between TAGLN expression and pathological characteristics of gastric cancer was also analyzed. Cell invasion and migration kits were used to further evaluate the effect of fibroblasts expressing high TAGLN (such as CAFs) on the metastasis of gastric cancer cell lines MKN45. Results No significant differences of the TAGLN mRNA and protein expression were detected among gastric cancer cell lines (P 〉 0.05). It was observed that the TAGLN expression in gastric cancer tissues was higher than that in the cancer-adjacent tissues ( P 〈 0.05). TAGLN was found to be expressed mainly in the stroma of gastric cancer by immunohistochemistry, such as fibroblasts, endothelial cells of new vessels or myocyte in tumor-invaded muscularis mucosae. Furthermore, compared with that in lymph node free group( 1. 084 ± 0. 328) , the TAGLN expression increased significantly in lymph node metastasis group( 3. 751 ± 0. 681 ) ( P 〈 0.05 ). The cell invasion and migration examination indicated that the CAFs expressing high TAGLN enhanced the invasion and migration ability of MKN45. However, when the TAGLN of CAFs was knocked down with siRNA, these cells lost the ability of faciliating the invasion and migration of MKN45. Conclusion The TAGLN is overexpressed in the stroma of gastic cancer, and it may enhance the cell invasion and metastasis ability of gastric cancer.
