摘　　要：目的了解SerpinB5在胃癌细胞株中的表达水平,探讨SerpinB5在胃癌发生发展中的作用。方法实时定量PCR检测胃癌细胞株及正常胃黏膜细胞株中SerpinB5的表达水平;化学合成小片断干扰RNA(siRNA)下调胃癌细胞株SerpinB5的表达;实时定量PCR及免疫印迹检测干扰前后SerpinB5mRNA及蛋白质表达的变化;MTT法、软琼脂克隆形成实验和体外Matrigel侵袭实验观察SerpinB5减敲后胃癌细胞株生物学行为的改变。结果SerpinB5在胃癌细胞株中的表达水平显著升高;经SerpinB5特异的siRNAmix作用后,SerpinB5mRNA及蛋白质表达明显下降;减敲SerpinB5后,胃癌细胞株HTB103的生长能力、克隆形成能力和侵袭能力均显著降低。结论SerpinB5的表达水平在胃癌细胞株中显著升高;SerpinB5在胃癌的发生发展中起促进作用。Objective To observe the expression level of SerpinB5 in gastric cancer cell lines and study the effect of SerpinB5 on the carcinogenesis and development of gastric cancer. Methods The expression levels of SerpinB5 in gastric cancer cell lines and non- transformed gastric cell lines were analysed by real-time PCR, The special siRNAs of SerpinB5 were obtained by chemosynthesis and transfected into gastric cancer cells, SerpinB5 mRNA and protein expressions were measured by real-time PCR and immunoblotting before and after the interference, and effects on biological behaviour of gastric cancer cell lines were also investigated by MTT colorimetry, soft agar clone formation assay and in vitro Matrigel assay, Results Compared with non-transformed gastric cell lines, the expression level of SerpinB5 in gastric cancer cell lines was up-regulated significantly. SerpinB5-specific siRNAmix led to decreased SerpinB5 mRNA and protein expressions. Knockdown of SerpinB5 in the HTB103 cell lines resulted in decreased cell proliferation, colony formation and invasion. Conclusion The expression level of SerpinB5 is significantly up-regulated in gastric cancer cell lines. SerpinB5 can promote the malignance of gastric cancer cell lines.
