替尼泊甙对胃癌细胞BGC-823体外增殖的影响

摘　　要：目的：观察替尼泊甙对胃癌细胞BGC-823体外增殖的影响。方法：取1.25mg/L,2.50mg/L,5.00mg/L,10.00mg/L,20.00mg/L的替尼泊甙作用于胃癌细胞BGC-82348h,用MTT法测定细胞生长抑制率;另取1.25mg/L的替尼泊甙作用于BGC-823细胞0h,12h,24h,48h,用流式细胞仪测定细胞凋亡率和细胞周期变化。结果：1.25-20.00mg/L的替尼泊甙对细胞生长的抑制率为15.99%-80.83%,细胞生长抑制率随替尼泊甙剂量的增加而升高;凋亡细胞随替尼泊甙作用时间的延长而增多,在48h细胞周期被阻滞在G2/M期。结论：替尼泊甙可抑制胃癌细胞的生长,其抑制作用存在剂量依赖性;凋亡细胞随替尼泊甙作用时间的延长而增加,其诱导凋亡的能力与细胞的G2/M期阻滞有关。Aim ： To investigate the effects of teniposide （ VM-26 ） on proliferation of gastric cancer cell line BGC-823. Methods：MTT assay was used to examine the inhibition rate of cell growth when cells were treated with 1.25 mg/L, 2.50 mg/L,5.00 mg/L, 10; 00 mg/L,20.00 mg/L VM-26 for 48 h; the apoptosis rate was examined by flow cytometry when cells were exposed to 1.25 mg/L VM-26 for 0 h, 12 h, 24 h, and 48 h. Results： The inhibition rate of cell growth was 15.99% - 80.83% when ceils were treated with 1.25 - 20.00 mg/L VM-26 and the inhibition rate increased in a dose-dependent manner; the apoptosis rate increased with exposure time. The cell cycle was arrested at G2/M phase at 48 h. Conclusion： The proliferation of BGC-823 cells could be inhibited by VM-26 in a dose-dependent manner; the apoptosis could be induced by VM-26, which may associate with VM-26 arresting cells at G2/M phase.
