广东地区宫颈癌HPV52型E6基因突变分析

摘　　要：目的调查我国广东地区宫颈癌患者组织中HPV52型的感染率及E6基因突变特性,为制备适合中国人群的疫苗提供流行病学资料。方法采用HPV通用引物对130例宫颈癌标本中的HPVL1区基因序列进行PCR扩增,并结合基因测序分析;对所获得的HPV52再进行E6基因测序分析。结果获得HPV52型8例,检出率为6.15%,对其中的3例HPV52E6基因进行测序分析。结论与GenBank标准株相比,广东地区发现的1例HPV52E6基因有多处突变,2例E6基因无突变,与香港发现的HPV52突变株相近。Objective To study the infectious rates and features of E6 genes of HPV52 in cervical cancer patients in population. Guangdong province and Methods DNA sampl provide epidemic data for development of HPV vaccine suitable for Chinese es of 130 cervical cancer tissues from patients in Guangdong province were amplified by HPV-specific PCR with consensus primers, then the E6 genes of HPV52 were sequenced for mutation analysis. Results Eight HPV52-positive cases were found in 130 cases and the infectious rate of HPV52 was 6.15%. E6 region nucleotide variation in 3 of the 8 cases were characterized by PCR-directed sequencing. Conclusion Compared with the HPV52 prototypes published in GenBank, several new point mutations at the E6 genes of HPV52 were found in only one case out of the three cases subjected to PCR-directed sequencing and these mutations are closed to those found in Hong Kong.
