摘 要:目的探讨上皮钙粘附蛋白(E-cadherin)和尿激酶型纤溶酶原激活剂(urokinase-type plasminogen activator,uPA)在脑膜瘤中的表达及临床意义。方法应用免疫组化SP法,对60例脑膜瘤手术根治标本进行E-cadherin和uPA测定。结果E-cadherin在10例正常脑组织中均呈强阳性表达,而在60例脑膜瘤中表达阳性率为69.2%,其差异有统计学意义(P<0·05)。但Ⅱ、Ⅲ级脑膜瘤与Ⅰ级脑膜瘤相比,E-cadherin的表达差异无统计学意义(P>0·05)。10例正常脑组织中uPA均呈阴性表达,60例脑膜瘤中uPA在Ⅱ、Ⅲ级脑膜瘤中的表达强度高于在Ⅰ级脑膜瘤中的表达,其差异有统计学意义(P<0·05)。E-cadherin低表达和uPA高表达与脑膜瘤侵袭性关系密切(P<0·05)。脑膜瘤E-cadherin和uPA蛋白表达呈负相关(P<0·05)。结论uPA可能在脑膜瘤的恶性进展中发挥某种作用;E-cadherin和uPA表达均是反映脑膜瘤的侵袭性重要指标,它们可能相互影响和共同调节脑膜瘤的侵袭性生长。Objective To investigate the expressions and significance of E-cadherin and uPA in human meningioma. Methods The expressions of E-cadherin and uPA were determined by the S-P immunohistochemical staining method in 60 cases of different pathological types of meningiomas. Results The positive rate of E-cadherin in normal brain tissue was 100% and in meningiomas was 69.2%. The difference between those was significant (P〈0. 05). The expression of E-cadherin in grade Ⅱ and grade Ⅲ meningiomas were not different from those in grade Ⅰ meningiomas(P〈0. 05). The expressions of uPA were significant higher in grade Ⅱ and grade Ⅲ meningiomas than those in grade Ⅰ meningiomas,in which the expression of uPA was significant higher than those in the normal human brain tissues(P〈0.05). Significant correlation was observed between low- expression of E-cadherin and high-expression of uPA with the invasion of the meningiomas. In addition there was a significant negative correlation between the expression of E-cadherin and uPA. Conclusion The expression of uPA possibly play a role in the malignant progression of human meningiomas. The expression of E-cadherin and uPA are important biological markers reflecting the invasiveness of meningiomas. They may exert mutual effect and co-regulate the invasive growth of human meningiomas.
