摘  要:目的观察心肺复苏术后大鼠海马神经元神经生长因子(nerve growth factor,NGF)、脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)表达的变化,以及APP17肽对NGF、BDNF表达的影响。方法采用窒息法制作大鼠呼吸心跳骤停模型,然后行心肺复苏术(cardiopulmonary re-suscitation,CPR)。♂性Wistar大鼠18只,随机分3组:手术对照组(A组)、心肺复苏组(B组)、心肺复苏+APP17肽组(C组),复苏至自主循环恢复(return of spontaneous circula-tion,ROSC)时静脉注射APP17肽10μg·(300g)-1。应用免疫组化方法观察各组大鼠复苏2h后海马神经元表达NGF、BDNF情况。结果与对照组(A组)相比心肺复苏组(B组)大鼠海马神经元NGF、BDNF表达明显降低(P<0.05);心肺复苏+APP17肽组(C组)NGF、BDNF表达明显增加,高于B组和A组(P<0.01)。结论心肺复苏模型大鼠自主循环恢复2h海马神经元NGF、BDNF表达降低,APP17肽能恢复神经元NGF、BDNF的表达,对缺血、缺氧性神经元损伤有保护作用。Aim To observe the effects of APP17 peptide on the changes in the levels of nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) in rat brain induced by cardiopulmonary resuscitation (CPR). Methods Cardiac-respiratory arrest model were made by apnea method, and followed by CPR treatment. Eighteen Wistar rats were randomly divided into three groups: Control group (Group A ), CPR group (Group B) and CPR plus APP17 peptide group (Group C), intravenous injection APP17 peptide when return of spontaneous circulation (ROSC). Expression of NGF and BDNF in hippocampal neurons were observed by immunohistochemisitry (IHC) method. Resuits The expression of NGF and BDNF in hippocampal neurons of rats in CPR group decreased significantly(P 〈0. 05); levels of NGF and BDNF in CPR plus APP17 peptide group were much higher than those in control group and CPR group(P 〈0. 01 ). Conclusion Expression of NGF and BDNF in hippocampal neurons decreases after circulatory recovery in CPR rats for two hours; APP17 peptide could increase expression of NGF and BDNF in hippocampal neurons and protect neurons from injury of hypoxemia and ischemia.
