摘 要:目的:研究联合应用百草枯(Paraquat,PQ)和代森锰(Maneb,MB)对帕金森病(Parkinson′s disease,PD)小鼠脑黑质神经胶质原纤维酸性蛋白质(Glial fibrillary acldic protein,GFAP)的表达变化。方法:PQ/MB建立C57BL/6J小鼠PD模型,利用免疫组化方法观察不同时间点小鼠黑质区酪氨酸羟化酶(Tyrosine hydroxylase,TH)和GFAP表达变化及相互关系。结果:模型组黑质致密部(Substantial nigra pars compacta,SNpc)区残留的TH阳性神经元呈进行性下降趋势,显著低于对照组的相应时间点(P<0.01),黑质区星形胶质细胞反应性增多并呈现明显活化状态。结论:PO/MB能诱导小鼠产生渐进性且不可逆的PD样改变,星形胶质细胞可能参与多巴胺能神经元的病理损伤过程。Objective: To investigate the expression of glial fibrillary acidic protein (GFAP) on parkinsonian C57BL/6J mice model induced by combination of paraquat and maneb. Methods:Twenty-four male mice were divided randomly into 2 groups: control group ( n = 10 ) and model group ( n = 14 ). As model for parkinson's disease, mice were injected for 6 weeks with paraquat and maneb. /mmunohistochemistry was used to measure the expression levels of tyrosine hydroxylase (TH) and glial fibrillary acidic protein (GFAP) in substantia nigra. Results:Mice injected with paraquat and maneb displayed marked behaviorial abnormalities, i.e., motor incoordination and a reduction of spontaneous motor behavior. The number of TH positive neurons in SNpc of model group decreased sharply after the last injection, and was lower than that of the control group ( P〈0.05 ). On the last time point, the TH positive neurons in model group decrease to 40% compared with control group. Since the 5th week after the last injection, mean gray value ( GV ) in model group (69.02 ± 1.25) was much higher than that of the control group ( 51.66±0.93, P〈0.05 ). Conclusions :Our results suggest that combination of paraquat and maneb could induce progressively and irreversibly changes in TH neurons of the substantia nigra pars compacta in mice, and astrocytes in substantia nigra may play an important role in the impairment of dopaminergic neurons in PD animal model induced by combination of paraquat and maneb.
