摘  要:目的:观察多种浓度丁酸、全反式维甲酸以及联合应用对肺腺癌细胞株A549β-连环素(β-catenin)及c-myc基因表达的影响。方法:用免疫组化法检测β-连环素(β-catenin)及c-myc基因表达情况。结果:经BA、ATRA处理72h后A549细胞β-cat、c-myc的AOD值降低,β-cat、c-myc蛋白阳性细胞比例减小,并呈剂量依赖性,其中BA和ATRA联合处理后较等浓度单药BA、ATRA作用更为显著。β-cat与c-myc的AOD值之间、β-cat与c-myc阳性细胞比例之间均分别呈正相关。结论:BA对肺腺癌A549细胞具有与ATRA相近的降低肺腺癌A549细胞β-cat、c-myc蛋白表达水平作用,并呈浓度依赖性,BA和ATRA联合应用具有良好的协同增效作用;BA、ATRA可能通过作用肺腺癌A549细胞Wnt信号通路影响细胞增殖。Objective: To observe the effects of butyric acid(BA), all-tram refinoic add(ATRA) on the expression of β- catenin and c-myc of human lung adenocarcinoma A549 cell line. Methods: The expression of theβ-catenin and c-myc genes were surveyed by immunohistochemistry assays. Results: Decreased AOD value, decreased positive cells percentage of β-cat and c-myc were found in the A549 cells exposed to BA (=Butyric acid) and ATRA (=All-trans refinoic add). After 72h's exposure, the two indexes decreased with a dose-dependent pattern. Even more prominent difference was found when A549 cells were ex- posed to BA and ATRA together after 72h. Meanwhile, linear correlation analysis demonstrated that β-cat had positive correlation with both the AOD and positive cells percentage of c-myc separately. Conclusions: Compared with ATRA, BA might play almost the same role to reduce the level of expression on-cat and c-myc of human lung adenocarcinoma A549 cell line in certain ranges, which was dependent on the dose,BA combined with ATRA could reinforce their effects each other on the A549 cell line, Further, BA and ATRA might affect the proliferation of the Human lung adenocarcinoma A549 cell line according to the Wnt signaling pathway.
