摘 要:应用人间充质干细胞作用肿瘤细胞,拟揭示间充质干细胞对肿瘤细胞的作用,为今后应用人间充质干细胞进行肿瘤细胞治疗奠定理论基础.应用胎儿真皮来源的 Z3间充质干细胞和胎儿骨髓来源的 BMMS-03间充质干细胞的条件培养液作用于乳腺癌 MCF-7细胞,观察 MCF-7细胞增殖和迁移能力的变化.结果显示,应用 BrdU 掺入分析和流式细胞仪测定等方法发现,在间充质干细胞条件培养液的作用下,MCF-7细胞的增殖受到了明显抑制;应用划痕实验和琼脂糖滴实验发现,MCF-7细胞的迁移能力也受到明显抑制.结果表明,人间充质干细胞对乳腺癌细胞的增殖和迁移具有抑制作用。To examine the effect of human mesenchymal stem cells (hMSCs) on human breast cancer MCF-7cells, the proliferation and migration of the cells were investigated. The hMSCs designated Z3 cells were established from human dermis tissues of a fetus aborted at 4 months gestation, immortalized by stable transfection of a pGRN145 plasmid containing the cloned hTERT gene. The MSCs designated BMMS-03 cells, derived from human fetal bone marrow in 4 month, were established without immortalization. In this study, we demonstrated that conditioned media from MSCs were able to decrease the proliferation and migration of MCF-7 cells in vitro. Proliferation of MCF-7 cells was decreased significantly by BrdU incorporation assay and flow cytometry assay after the treatment. Moreover, wound healing assay and agrose drop assay showed that the migration of MCF-7 cells was inhibited as well. Our finding demonstrated that hMSCs can directly inhibit breast cancer MCF-7 cells in vitro. Therefore, we conclude that hMSCs have a potential role in controlling proliferation and migration of solid tumor cells.
