重组人甲状旁腺激素(1-34)在体外对UMR106细胞增殖、分化功能的影响

摘　　要：目的:观察重组人甲状旁腺激素(1-34)[recombinant human parathyroid hormone(1-34),rhPTH(1-34)]对成骨样细胞 UMR106细胞增殖、分化功能的影响,初步探讨其作用机理。方法:采用体外培养的 UMR106细胞,在连续或间歇2种给药方式下,以1×10^(-12)～1×10^(-7)mol·L^(-1)rhPTH(1-34)作用于细胞,选择 MTT 法考察 rhPTH(1-34)对成骨细胞分裂和增殖的影响,同时测定细胞裂解液中代表成骨细胞分化功能的碱性磷酸酶活性。结果:rhPTH(1-34)连续作用于 UMR106细胞时,能抑制增殖和分化,且浓度越高,抑制作用越强;间歇作用于 UMR106细胞时,能够促进细胞的增殖和碱性磷酸酶活性的升高,并且浓度为1×10^(-10)mol·L^(-1)时,促进作用最强,而高于或低于该浓度,作用减弱。结论:rhPTH(1-34)对 UMR106细胞增殖和分化功能的影响与给药方式和药物浓度有关。连续给药时,抑制 UMR106成骨样细胞的增殖和分化,浓度越高,抑制作用越强;而间歇给药时,促进成骨样细胞的增殖和分化,其剂量反应曲线呈类似钟形曲线。Objective ： To observe the effect of recombinant human parathyroid hormone （ 1 - 34） [ rhPTH （ 1 - 34） ] on proliferation and differentiation of UMR106 osteoblast - like cells in vitro. Method ：In vitro culture systems, while given with 1×10^12 to 1×10～mol·L^-1 rhPTH（ 1 - 34） continuously or intermittently,the proliferation and differentiation of UMR106 osteoblast -like cells was examined by MTr assay and ALP activity. Result：When osteoblastic cells were exposed to rhPTH （ 1 - 34） continuously, the agent inhibited osteoblast proliferation and differentiation, but when the cells were treated intermittently with rhPTH （ 1 - 34）, it exerted an anabolic effect on proliferation and dif- ferentiation of UMR106 osteoblastlike cells depending on the concentration of rhPTH（ 1 -34 ）. Conclusion： Resuits of the experiments in vitro indicated that rhPTH （ 1 - 34） has diverse effects on osteoblast - like cells depending on the mode and concentration of administration.
