摘 要:目的探讨同型半胱氨酸(Hcy)与谷氨酸钠(MSG)联合作用,建立孕鼠妊娠期高血压疾病模型的可行性。方法将成年妊娠 Wistar 雌鼠随机分为4组,每组10只。从孕第10天起,对照(PN)组和谷氨酸(PG)组孕鼠分别每日腹腔内注射生理盐水2ml,Hcy(PH)组、Hcy+PG(PHG)组孕鼠分别每日腹腔内注射Hcy 200 mg/kg;PN、PH 组孕鼠隔日背部皮下注射生理盐水2 ml,PG、PHG 组孕鼠隔日背部皮下注射 MSG 1 g/kg;直至分娩。孕期测定大鼠血压、尿蛋白、肝肾功能及行为变化;分娩后测量胎盘湿重、仔鼠数量、仔鼠体重、身长,观察大脑皮层、肾脏及胸主动脉组织结构改变。结果 (1)PH、PHG 组孕鼠于孕第12天起,出现血压升高,分别为(107±8)、(109±10)mm Hg(1 mm Hg=0.133 kPa),至孕第20天达最高峰,分别为(119±10)、(121±7)mm Hg,与 PN、PG 组比较,差异均有统计学意义(P<0.01);(2)PH、PHG 组孕鼠孕第15天时,尿蛋白含量分别为(1.42±0.53)、(1.53±0.24)g/L,与 PN、PG 组比较,差异也有统计学意义(P<0.01);(3)PH、PHG 组丙氨酸转氨酶(ALT)分别为(57±15)、(69±24)U/L,天冬氨酸转氨酶(AST)分别为(265±61)、(293±118)U/L,尿素氮(BUN)分别为(9.5±0.8)、(9.5±1.6)mmol/L,肌酐(Cr)分别为(54±10)、(54±10)μmol/L,与 PN、PG 组分别比较,差异均有统计学意义(P<0.05);(4)PH、PHG 组仔鼠体重分别为(3.5±3.9)、(3.3±3.7)g,胎盘湿重分别为(0.49±0.28)、(0.45±0.03)g、仔鼠身长分别为(3.6±1.5)、(3.5±1.5)cm。分别与 PN、PG 组比较,差异也均有统计学意义(P<0.01);(5)光镜下,PHG 组孕鼠出现肾小球基底膜增厚,系膜细胞增生,肾小管上皮细胞水肿;大脑皮层神经细胞出现固缩性退行性变;电镜下,PHG 组孕鼠出现主动脉内皮细胞细胞小器缺乏,中膜平滑肌细胞增生;(6)PHG 组孕鼠可出现明显的行为改变,如阵发性凝视、动须、面部抽搐、一侧前肢震颤等。结论Hcy 与 MSG 可联合作用,通过损伤血管内皮细胞及大脑皮层�Objective To determine whether homocysteine (Hey) and monosodium glutamate (MSG) could lead to animal model of hypertensive disorder complicating pregnancy and its mechanism. Methods Female adult Wistar rats were randomly divided into 4 groups: pregnant control group ( PN ), pregnant Hey group (PH) , pregnant glutamic acid group (PG) and pregnant Hey and glutamic acid group (PHG). The rats of each group were injected with Hey 200 mg/kg or physiological saline every day intraperitoneaUy and with MSG or 0. 9% saline every other clay via Hey injection from the 10th day to the 20th day of pregnancy. The blood pressure, urine protein, function of liver and kidney, weight of placenta, length and weight of fetus were all measured. The histological change of the pallium and the change of behavior of pregnant rats were also observed. Results ( 1 ) The blood pressure in PH[ ( 107 ± 8 ) mm Hg, 1 nun Hg =0. 133 kPa], and PHG group [ ( 109 ± 10) mm Hg] after the treatment increased significantly compared with those in other groups from the 12 th day after pregnancy(P 〈0. 01 ). (2) The level of urine protein [ ( 1.42 ± 0. 53 ) g/L, ( 1.53 ± 0. 24 ) g/L ] in PH and PHG groups after the treatment was significantly higher than the other groups(P 〈0. 01). (3)Obvious changes of the function of liver and kidney [alanine aminotransferase (ALT) ( 57 ± 15 ) U/L, ( 69 ± 24 ) U/L, aspartate aminotransferase (AST) ( 265 ± 61 ) U/L, (293 ± 118 ) U/L, blood urea nitrogen (BUN) ( 9.5 ± 0. 8 ) mmol/L, ( 9.5 ± 1.6 ) mmol/L, ereatinine (Cr) (54 ± 10)μmol/L,(54 ± 10)μmol/L] ,were found in PHG group and PH group(P 〈0.01). (4) The weight of placenta [ (0. 49 ± 0. 28 ) g, (0. 45 ± 0. 03 ) g], length and weight of fetus [ ( 3.6 ± 1.5 ) cm, ( 3. 5 ± 1.5) cm ] in PH group and PHG group were significantly lower than that in PN group and PG group ( P 〈 0.01 ).
