摘 要:目的 观察地塞米松(DEX)对内毒素(LPS)诱导的肾小球系膜细胞(MC)中细胞因子的产生及其基因表达的影响。方法 采用免疫学和分子生物学方法。结果 1.细胞因子在正常MC中分泌量极微,其mRNA表达处于低水平。2.LPS诱导MC细胞因子的产生及mRNA表达增加。3.DEX对细胞因子的产生及mRNA表达有明显的抑制作用,呈剂量依赖关系。结论 DEX是MC产生细胞因子的较强抑制剂,为今后临床上治疗肾小球肾炎,提供了理论及实验依据。Objective To study the effects of dexamethasone (DEX) on the cytokines production and gene expression in the cultured glomerular mesangial cells (MO induced by lipopolysaccharide (LPS). Methods The cytokines and gene expression were detected by using immunological method and molecularbiological method. Results 1. The cytokines production and the mRNA expression of cytokines in normal MC were at a low level. 2. The protein production and mRNA expression of cytokines induced by LPS increased. 3. DEX showed an inhibition on the cytokines production and mRNA expression induced by LPS. A dose dependent correlation were found. Conclusion DEX is an inhibitor on cytokines production and mRNA expression in MC. This provides an experimental evidence for clinical treatment in glomerulonephritis.
