摘 要:目的:探讨一氧化氮NO(Nitric Oxide,NO)对成骨细胞前体细胞系MC3T3-E1的增殖和矿化调控作用。方法:给于成骨细胞样细胞浓度为1×10-3mmol/L的NO外源性供体S亚硝基N乙酰基青霉胺(S-nitroso-N-acetyl-dl-penicillamine,SNAP)3w,通过成骨细胞DNA含量及碱性磷酸酶(Alkaline Phosphatase,ALP)活性变化,以检测NO对细胞生长影响,通过Von Kossa染色和骨钙素免疫组化方法,以观察成骨细胞合成钙盐情况;通过Western Blot方法检测细胞骨唾液酸蛋白(Bone Sialoprotein,BSP)表达变化。结果:和对照组相比,SNAP可明显增加成骨细胞DNA含量和碱性磷酸酶活性;SNAP可刺激成骨细胞于培养2w时生成明显钙化结节,骨钙素蛋白表达,并显著上调骨唾液酸蛋白表达。结论:一定浓度一氧化氮,可加速成骨细胞前体细胞MC3T3-E1增殖,并可能通过调控骨唾液酸蛋白表达刺激成骨细胞进一步分化和矿化。Objective: To study the effect of nitric oxide on cell proliferation, differentiation and biomineralization of osteoblast-like MC3T3-E1. Methods: 1 × 10^-3 mmol/L NO donor administered to MC3T3-E1 cells for three weeks. The effect of SNAP content and alkaline phosphatase (ALP) activity, and biomineralization S-nitroso-N-acetyl-dl-penicillamine (SNAP) was on cell proliferation was valuated through DNA was determined through calcium deposit by Von Kossa Assay, osteocalcin expression was examined by immunohistochemistry, and bone sialoprotein (BSP) expression by Western blot. Results: Compared with the control group, SNAP significantly raised the DNA content and ALP activity in osteoblast-like cells with the existence of β -glycerolphosphate sodium and ascorbic acid. In addition, SNAP promoted formation of calcium nodule and the expression of osteocalcin, the expression of BSP was also up-regulated after two weeks' culture. Conclusion: NO may stimulate the proliferation of MC3T3-E1 cells, regulate the differentiation and biomineralization through the expression of BSP.
