高效液相色谱法测定铁苋菜中没食子酸含量

摘　　要：目的测定铁苋菜中没食子酸的含量。方法采用高效液相色谱法,Zorbax-C18色谱柱,流动相为水-二甲基甲酰胺-冰醋酸(99.82∶0.15∶0.03),检测波长为270nm,流速为1mL/min,柱温为室温。结果没食子酸进样量在0.081～0.811μg范围内与峰面积呈良好的线性关系(r=0.9999),平均日内、日间RSD为1.01%和1.60%(n=6),不同浓度的平均回收率为97.68%,RSD=1.43%(n=6)。结论该法快速简便,稳定可靠,专属性强。Objective To determine the content of gallic acid in Acalypha australis L. with HPLC, Methods A zorbax C18 column （250 mm × 4, 6 mm, 5 μm） was used. The mobile phase was water - dimethyformamide - glacial acetic acid （99. 82 ： 0. 15 ： 0. 03）. The detection wavelength was at 270 nm with flow rate of 1.0 mL/min, and the temperature of column was at room temperature. Results The linearity of gallic acid was over the range of 0. 081 -0. 811 p.g/mL with the correlation coefficient 0, 999 9. The average recovery rate of different concentrations was 97.68% with RSD 1.43% （n=6）.The within-day and between-day RSD was 1.01% and 1.60% （n=6）.Conclusion The method is convenient, stable, reliable and special for the dermination of gallic acid in Acalypha australis L..
