摘　　要：目的研究HBV基因在已传20代BALB／c-TgN（HBV D型1．3）SWS458小鼠中复制和表达情况。方法采用PCR荧光定量检测法、ELISA检测和免疫组化的方法研究HBV基因在转基因小鼠体内的复制和表达情况。结果F20代BALB／c-TgN（HBVD型1．3）SWB458小鼠血清HBVDNA为（104—106）copy／mL，HBsAg表达量为（124．41±33．46）ng/mL，preS1和HBcAg的OD值分别为0．248±0．076和0．635±0．338。肝组织有HBsAg和HBcAg表达，且HBsAg为胞浆型，HBcAg为核型。结论经过筛选和培育，本转基因小鼠传至20代时HBV基因复制表达稳定，表明我们已经获得一个传代表达稳定的HBV转基因小鼠品系。Objective To evaluate replication and expression of HBV genome in the F20 generation BALB/c-TgN（ HBV D 1.3）SWB458. Methods Replication and expression of HBV genome was estimated by RT-PCR, ELISA and immunohistocbemistry. Results Serum HBV DNA content of the F20 generation BALB/c-TgN（ HBV D 1.3） SWB was （104 - 106）copy/mL. The expression level of HBsAg was （124.41± 33.46）ng/mL and the OD value of pre S1 and HBcAg were 0.248 ±0.076,0.635±0.338 respectively. HBsAg and HBcAg were expressed in the liver. Furthermore, HBsAg was dectable mainly in the cytoplasm while HBcAg was mainly present in the nucleus. Conclusion This results demonstrate that the HBV genome can be stably transmitted until F20 generation and replicated and expressed in the F20 generation through unceasing screening and breeding. This indicates that we have obtained a HBV transgenic mice strain in which HBV genome can be transmitted and expression stably.
