摘　　要：目的探求0-氯乙酰-氨甲酰基烟曲霉醇(TNP-470)联合高聚生(HAS)治疗恶性腹水的有效性。方法制造小鼠恶性腹水模型(40只),将其随机分为4组:0.9%NaCl对照组(A组)、TNP-470治疗组(B组)、HAS治疗组(C组)、TNP-470联合HAS治疗组(D组),分别腹腔内注射给药;观察4组的生长情况、存活时间、腹水量;利用免疫组化方法检测各组腹膜上肿瘤组织微血管密度(MVD),利用Tunnel方法检测腹水中肿瘤细胞的凋亡率,利用ELISA方法检测小鼠血清中IFN-γ浓度。结果D组小鼠生命延长,腹腔肿瘤细胞的凋亡率(AIs)明显升高,与其他组比较,差异有显著意义(P<0.01)。D组血清中肿瘤坏死因子(IFN-γ)的浓度明显升高,与其他各组比较,差异亦有显著意义(P<0.05)。TNP-470联合HAS治疗使腹膜上的微血管密度显著降低,与其他组比较,差异有显著意义(P<0.01),并显著地抑制了恶性腹水的产生。结论TNP-470联合HAS是治疗恶性腹水的有效方法。Objective To evaluate the efficacy of therapy combined TNP-470 with highly agglutinative staphylococcin （HAS） in treatment of malignant ascites in mice model. Methods 40 mice with malignant ascites established were randomly divided into 4 groups, and received intraabdominal injection with 0. 9% NaCl （control group, group A）, TNP-470 （group B）,HAS （group C）, TNP-470 plus HAS （group D）. Their growth, survival and malignant ascites were monitored;MVD in tumors was detected by immunohistochemistry and apoptosis of tumor cells was evaluated by Tunnel. The lever of IFN-γ in serum was determined with ELISA. Results The level of IFN-γ in serum and the apoptosis of tumour cell were increased significantly （P〈0.01） of the mice in group D, and the survival of mice in group D was the longest among the 4 groups. The MVD was the less and the growth suppression of the established malignant ascites was found in group D. Combination therapy significantly prolongs the survival of mice bearing tumor. Conclusion The therapy combined TNP-470 with HAS for malignant ascites significantly warrants investigation as therapeutic strategy.
