摘 要:通过液体培养和HPGFC测定,筛选获得有效成分和糖肽产量均较高的发酵生产菌株LS,然后优化其发酵培养基,并在30L发酵罐中进行放大实验。结果表明,在30L发酵罐中,发酵96h,菌体干重为24.29g/L,糖肽产量可达1.63g/L,经测定,提取所得的胞内糖肽其出峰时间与对照品基本一致,其蛋白质含量、总糖含量和单糖含量均符合国家药品标准WS-XG-021-2002要求,为云芝糖肽的工业化发酵生产奠定了基础。By liquid culture and high performance gel filter chromatography(HPGFC), Coriolus versicolor strain LS with higher active component and higher poiysaccharopeptide yield was selected for polysaccharopeptide production by submerged fermentation, After optimization of fermentation medium, the fermentation in 30L fermentor by Coriolus versicolor strain LS was carried out. The experimental results showed that the maxium biomass was 24.29g/L after cultivation for 96 hours. The yield of polysaccharopeptide was 1.63 g/L. HPGFC retention time of extracted polysaccharopeptide was consistent with that of standard, Its total sugar content, protein content, and reducing sugar content were in accordance with China medicine standard WS-XG-021-2002, The foundation for industrialization fermentation of Coriolus versicolor polysaccharopeptide was provided.
