摘 要:目的:探讨口腔鳞癌组织中细胞角蛋白19(CK19)的蛋白质表达与基因转录之间的关系及其临床意义。方法:采用免疫组织化学和荧光实时定量RT-PCR方法,对33例口腔鳞癌患者的癌组织及癌旁组织中CK19的蛋白质表达和mRNA水平进行检测,采用SPSS10.0软件包对数据进行配对t检验和相关分析。结果:口腔鳞癌组织中,CK19蛋白质和mRNA表达呈正相关,且均显著高于癌旁组织。癌组织CK19蛋白质表达阳性率为90.91%(30/33),mRNA水平是癌旁组织的2.21倍。CK19蛋白质和mRNA表达均与肿瘤的恶性程度显著相关。恶性程度越高,两者的表达水平也越高。结论:口腔鳞癌组织中CK19蛋白质和mRNA表达上升,并且与肿瘤恶性程度相关,CK19免疫组织化学和荧光实时定量RT-PCR技术在鉴别组织恶性程度上具有潜在的临床价值。PURPOSE: To investigate the protein expression arid gene transcript level of cytokeratin 19 (CK19) in oral squamous cell carcinoma tissues as well as their correlation and clinical significance, METHODS: 33 oral squamous cell carcinoma patients confirmed by pathologic diagnosis were involved in this study, paired samples of the cancerous and para-cancerous tissues from the same patients were used for the detection of CK19 protein expression and CK19 mRNA level by immunohistochemistry and fluorescent quantitive real-time RT-PCR assays respectively, The data was analyzed by SPSS10,0 software package for paired t test and correlation analysis, RESELTS: Among the 33 oral squamous cell carcinoma patients, there were 28 males and 5 females, their age ranged from 34 to 78 years with a mean age of 58.8 years, The CK19 protein expression and mRNA transcript level in cancerous tissues increased significantly compared with the para-cancerous tissues, there was significant positive correlation between the CK19 protein expression and mRNA transcript level. The positive rate of CK19 protein expression in cancerous tissue was 90.91% (30/33), and the mRNA level in cancerous tissue was 2.21 folds as that in the para-cancerous tissue. Both of the CK19 protein expression and CK19 mRNA level in cancerous tissue correlated significantly with the malignant degree of tumors; the poorer the pathological differentiation degree, the stronger and higher level of CK19 protein expression and mRNA level. CONCLUSION: CK19 protein expression and CK19 mRNA level increase in oral squamous cell carcinoma tissues with strong correlation with the malignant degree of tumor. The potential differential value is suggested on the detection of CK19 protein expression and mRNA level in cancerous tissues by immunohistochemistry and fluorescent quantitive real-time RT-PCR techniques.
