摘  要:目的探讨小鼠神经干细胞(NSCs)在体外和体内对胶质瘤细胞的趋向性。方法将原代培养6d的小鼠NSCs悬液离心、收集备用。制备C6胶质瘤细胞、3T3成纤维细胞爬片,随后与NSCs共同培养3d。用荧光染料Hoechst33342标记NSCs,借助立体定向技术于胶质瘤动物模型内接种NSCs,观察NSCs在胶质瘤动物模型脑内的分布情况。结果小鼠NSCs对C6细胞有明显的趋向性。体内实验见肿瘤组织内满布标记的NSCs。结论小鼠NSCs无论在体外或体内对胶质瘤细胞均有一定的趋向性。Objective To explore the tropism of mouse neural stem cells (NSCs) for C6 glioma cells cultured in vitro and in vivo. Methods The NSCs cultured for 6 d in vitro were centrifuged to reserve, and C6 cells patch and 313 fibroblasts patch were prepared at the same time. NSCs were co-cultured with C6 cells on patch for 72 h, while the 313 fibroblasts patch was used as control group. To investigate the tropism of NSCs for glioma tissue in vivo, rat C6 glioma model was established by stereotactic technique. The NSCs tagged with fluoresent Hoechst33342 were implanted into the location of glioma. After 5 d of implantation, sections from rat C6 glioma models were analyzed, Results The number of nem'espheres suspended moving to the C6 cells patch was more than that in 313 fibroblast patch. The density of NSCs in glioma tissue was larger than that in the surrounding normal brain tissues. Condusion NSCs have a tropism for glioma either in vivo or in vitro.
