摘  要:【目的】观察兔眼视网膜细胞经瞳孔温热疗法(TTT)不同能级照射后细胞凋亡情况。【方法】选择健康青紫兰兔24只,对实验眼采用1.2 mm光斑的810 nm激光照射60 s。各组照射剂量分别为阈能量的120%,100%,80%,50%。采用原位缺口末端标记法(TUNEL)、荧光素标记的连接素/碘化丙碇(AnnexinⅤ-FITC/PI)双染色流式细胞测定法观察视网膜细胞的凋亡。【结果】各能级均有细胞凋亡的发生,以内颗粒层为主,以80%能级为多。照射后早期以凋亡为主,随时间延长,光照后24 h,凋亡继发坏死增加。【结论】低能级照射以细胞凋亡为主,高能级照射以细胞坏死为主,80%能级为较安全能级。[Objective]To observe the apoptosis of rabbit's retinal cells induced by TTT (transpupillary thermotherapy). [Methods]TTT was performed on eyes of 24 young healthy pigmented rabbits with 1.2 mm spot size and 60s irradiation. 120 %, 100%, 80 %, 50% of threshold power were used for each group. Apoptotic cells were detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling, Annexin V-fluorescein isothiocyanate/ Propidium iodide labeling and flow cytometry. [Results]Apoptotic cells could be detected in all fundus layers (mostly in inner nuclear layer) at all levels (especially in 80% level). There were mostly apoptotic cells in early period of postirradiation ,and with time going (24th hour after postirradiation), cellular apoptotic necrosis continued with apoptosis was increasing. [Conclusion]There are more apoptotic cells observed in lower power levels, and more necrosis cells observed in higher power levels, 80% of threshold power is the safer level.
