摘  要:目的检测人上皮性卵巢癌细胞株SKOV3移植裸鼠腹腔成瘤后血管生成拟态的形成和分子塑形的改变。方法采用慢病毒转染SKOV3细胞携带绿色荧光蛋白,接种裸鼠腹腔成瘤后,苏木精-伊红染色、透射电镜、荧光显微镜观察肿瘤组织切片,免疫组化、肿瘤原代培养后采用免疫荧光化学、流式细胞仪、RT-PCR等技术检测SKOV3细胞是否表达血管内皮标记CD31等。结果苏木精-伊红染色、透射电镜、荧光显微镜证实SK- OV3细胞裸鼠腹腔内成瘤后形成了血管生成拟态,即SKOV3细胞在没有宿主血管内皮细胞参与下,形成特有的腔隙样管道,内有红细胞衬覆。在原代培养的SKOV3细胞,用免疫荧光化学、RT-PCR检测显示SKOV3细胞表达CD31,用流式细胞仪检测,SKOV3细胞表达CD31、Ⅷ因子,而不表达VEGF。结论人上皮性卵巢癌细胞株SKOV3在体内能表达部分血管内皮细胞的特异性抗原,具有可塑性,可在体内形成血管生成拟态。To determine the plasticity of human epithelial ovarian cancer cell SKOV3 and formation of vasculogenic mimicry in vivo. Methods Lentiviral vector carrying green fluorescence protein was transduced SKOV3. SKOV3 cells carrying green fluorescence protein were implanted into peritoneal cavity of nude mice. When the transplantable tumor develop, section of paraffin-embeded, formaldehyde-fixed tissues were sectioned and stained with H-E. Tumor tissue was also studied by electron microscopy and fluorescence microscopy. Expression of vascular cell marker such as CD31 was detected by immunoehemical staining and immunofluores- cent method,flow cytornetry and RT-PCR after primary cell culture. Results H-E staining,electron microscopy and fluorescence microscopy indicated that SKOV3 engaged in vasculogenic mimicry in vivo. SKOV3 formed patterned networks with red blood cells inside. After the tumor cells were primary cultured, immunofluorescent method and RT-PCR indicated that CD31 mRNA was expressed in SKOV3 cells, while flow cytometric analysis showed that SKOV3 expressed CD31 and Ⅷ factor but failed to express VEGF. Conclusions Human ovarian cancer cell SKOV3 could express some particular antigen of vascular epithelial cells in vivo. Formation of vasculogenic mimicry in vivo reflects the plasticity of SKOV3.
