摘 要:目的探讨膜组织结构伸展刺突蛋白(简称膜突蛋白)在喉癌和转移淋巴结中的表达及其与临床各相关因素之间的关系。方法采用逆转录聚合酶链反应(reversetranscriptasepolymerasechainreaction,RT-PCR)技术和免疫组织化学(immunohi-stochemistry)检测膜突蛋白在58例喉癌组织、癌周非癌组织和25例喉癌转移淋巴结中的表达。结果RT-PCR结果显示膜突蛋白在癌周上皮组织、喉癌和转移淋巴结中的表达分别为0.086±0.063、0.599±0.301和0.731±0.096,各组表达量之间差异显著,具有统计学意义(F=132.456,P<0.001);免疫组化显示膜突蛋白在蛋白水平发现膜突蛋白在癌周上皮组织、癌组织和颈部转移淋巴结中的表达依次增高(P<0.05),其表达与临床分期、T分级、颈淋巴结转移和病理学分级具有相关性(P<0.05),但与患者性别、年龄以及吸烟程度无相关性(P>0.05)。结论膜突蛋白是细胞膜与肌动蛋白丝之间连接的桥梁,参与细胞表面的构建及伪足的形成,可能在喉癌的侵袭和转移过程中发挥重要的作用。OBJECTIVE To evaluate the expression of moesin (membrane-organizing extension spike protein)in laryngeal carcinoma and its related clinical factors. METHODS RT-PCR (reverse transcription polymerase chain reaction) and immunohistochemistry methods were used to investigate the expression of moesin in normal epithelium tissues adjacent to cancer, laryngeal carcinoma tissues in 58 patients and in metastastic lymph nodes in 25 patients. RESULTS The result of RT-PCR showed that the expression of moesin in normal epithelium tissues adjacent to cancer(0.086+0.063), laryngeal carcinomas(0.599± 0.301) and metastastic lymph nodes(0.731±0.096)had significant difference (F=132.456, P〈0.001). The results of immunohistochemistry showed that the expression of moesin gradually increased in normal epithelium tissue adjacent to cancer, laryngeal cancers and metastastic lymph nodes(P〈0.05), which is related with the clinical stages, T grading, the metastasis of cervical glands and pathology grading(P〈0.05), but no relation with gender, age and smoking(P〉0.05). CONCLUSION Moesin is a membrane/actin filament protein involved in dynamic restructuring of the cell surface and filopodia, a cell structure needed for cell adhesion and motility, which may play an important role in the process of invasion and metastasis of laryngeal carcinoma.
