益气解毒颗粒对鼻咽癌细胞株HNE1凋亡诱导活力的研究

摘　　要：目的从益气解毒颗粒对体外培养的人鼻咽癌细胞株HNE1的细胞凋亡诱导活力角度探讨该方的抑癌机制。方法采用流式细胞术和免疫细胞化学法比较观察益气解毒颗粒原方及其拆方药物血清对体外培养的HNE1细胞凋亡诱导活力差异。结果益气解毒颗粒原方15%药物血清对HNE1的细胞凋亡诱导效应最为明显,48小时显示细胞凋亡指数13.69±0.26,且具有时间和药物浓度双重依赖性的特点,而在增强p53表达效应上,则以原方和益气组最为明显。结论益气解毒颗粒诱导鼻咽癌细胞凋亡主要是通过其益气组的功效而得以发挥,其机制之一可能是通过对p53的表达上调而实现的。OBJECTIVE To investigate the effects of qi-boosting toxin-resolving formula （QBTRF） on the apoptosis in nasopharyngeal carcinoma cell line（ HNE1）. METHODS All samples of medicinal serum were cultured together with HNE1 in vitro to investigate their effects on apoptosis in HNE1 cells with flow cytometry and immunocytochemistry. RESULTS All fractions of medicinal serum could induce apoptosis in HNE1 cells at the concentration of 15%, especially for the composition of original formula. Its apoptotic index at 48h was 13.69±0.26. These effects showed both characters being time-dependent and concentrationdependent. All fractions of medicinal serum were able to promote the expression of p53, especially for group Ⅰand Ⅱ. CONCLUSION QBTRF may bring about its effects on target cells to induce apoptosis through regulating the expression of associated genes. The effect is related with the fractions of Qi-boosting expression.
