摘 要:高质量的蛋白样品制备是进行双向电泳的前提条件。杨树叶片中因含有丰富的多酚、色素、多糖等物质,给蛋白质的提取带来了困难。为了找到一种适合杨树叶片的蛋白质提取方法,以南林895杨为实验材料,采用三氯乙酸-丙酮沉淀法对杨树叶片蛋白质进行提取。对所提取的蛋白分别用考马斯亮蓝染色法和银染法进行检测,得到了良好的双向电泳图谱。High-quality protein sample preparation is the premise of two-dimensional gel electrophoresis (2-DE). Poplar leaves contain abundant interfering compounds such as polyphenols, pigments, polysaccharides and so on order to find a method suitable for protein ex was used as the experimental material. The was adopted to extract proteins. Coomassie used respectively to detect the extracted p paper provides a basis for poplar proteom rot ics e , which make traction of po method of tri brilliant blue ins. Good pro it difficult to extract proteins. In plar leaves, the poplar of NL895 chloroacetic acid (TCA)-acetong was adopted to extract proteins.Coomassie brilliant (CBB) and silver staining were used respectively to detect the extracted proteins.Good protein patterns were obtained. This paper provides a basis for poplar research in our lab.
