丹参酮Ⅱ_A对小鼠骨髓瘤细胞SP2/0体外增殖与凋亡的影响

摘　　要：目的研究丹参酮ⅡA(TanshinoneⅡA,TanⅡA)对小鼠骨髓瘤细胞SP2/0的生长抑制与诱导凋亡作用。方法选用小鼠骨髓瘤细胞系SP2/0,运用光镜、MTT法、DNA琼脂糖凝胶电泳分析SP2/0细胞增殖及凋亡的改变。结果经丹参酮ⅡA处理后,SP2/0细胞增殖明显被抑制,当4.0μg/mlTanⅡA处理SP2/0细胞72小时,增殖抑制率达(79.6±2.1),镜下可见凋亡细胞的形态学改变,细胞DNA片断化(呈现“梯状”断裂现象)。结论TanⅡA能有效地抑制小鼠骨髓瘤细胞系SP2/0增殖,促进SP2/0细胞凋亡。Objective To study the effect of Tanshinone Ⅱ A on the proliferation, apoptosis of mouse cell line SP2/0 in vitro. Methods The cell proliferation was measured by cell counting, MTF assay. Cell apoptosis was determined by morphology, agarose gel electrophoresis methods. Results After treatment with 0.25 -4.0μg/ml Tan Ⅱ A for 72h, the proliferation of SP2/0 cells was significantly suppressed, and the morphologic characteristics of apoptotic cells were observed under microscope, DNA ladder was presented in DNA electrophoresis. Conclusion Tan Ⅱ A can effectively inhibit the proliferation and induce the apoptosis of mouse SP2/0 cell line in vitro.
