摘 要:目的观察不同强度微波辐射对视网膜神经节细胞(retinal ganglion cells,RGCs)凋亡的影响。方法体外培养RGCs,经频率为2450MHz微波辐射1h,按辐射强度分为4组:0mW·cm-2组、10mW·cm-2组、30mW·cm-2组和60mW·cm-2组。倒置显微镜下观察细胞形态,台盼蓝染色检测细胞存活率,Annexin V-PI荧光标记双染色流式细胞术和TUNEL法检测RGCs的凋亡率。结果各组细胞经辐射后形态均有所改变,细胞存活率分别为98.7%、98.4%、87.8%和63.8%,随微波强度增加而下降;而流式细胞术检测的细胞凋亡率的结果表现出强度及时间相关性,TUNEL法检测的细胞凋亡率分别为5.2%、12.2%、18.0%和18.5%,随微波强度增加而增加。结论2450MHz微波可引起RGCs损伤及细胞凋亡,其损伤程度表现出辐射剂量相关性。Objective To observe the effect of different intensity of microwave irradiation on retinal ganglion cells (RGCs). Methods RGCs were cultured in vitro and exposed to 2 450 MHz microwave for 1 hour. Based on different intensity of microwave irradiation,the cells were divided into four groups:0 mW · cm^-2 group,10 mW · cm^-2 group,30 mW · cm^-2 group and 60 mW · cm^-2 group. The morphological variation of cells was observed by invert microscope. The survival rate was detected by trypan blue. Annexin V-PI two-colour flow cytometry and TUNEL assay were used to detect the percentage of apoptotic cells after radiation. Results The changes of morphology of all cells were observed after radiation. The survival rates were 98.7% ,98.4% ,87.8% and 63.8% respectively,which decreased with the increase of microwave intensity,but the percentages of apoptotic cells increased in a dose- and time-dependent manner by flow cytometry,and which were 5.2% ,12.2% ,18.0% and 18.5% respectively by TUNEL assay. Conclusion 2 450 MHz microwave can induce the damage and apoptosis of RGCs in a dose-dependent manner.
