摘  要:通过反转录-聚合酶链反应(RT-PCR)扩增出了中国鹿狂犬病野毒株(8202株)糖蛋白(G)基因膜外主要功能区的2个片段,共约800个bp(370~1179),含有可以诱导中和抗体的2个抗原决定簇的基因片段和决定毒力及与神经细胞受体结合部位的基因片段。通过平端连接将2个片段克隆到pUC18的SmaⅠ位点,采用Sanger双脱氧终止法测定cDNA片段的核苷酸序列,并推导出其氨基酸序列。将这一序列与国内外已发表的11株狂犬病病毒的相应序列输入计算机进行比较分析,结果表明8202株主要功能区与上述11个毒株核苷酸序列的同源性在73.0%~96.4%之间,氨基酸序列的同源性在86.4%~94.4%之间,8202株与中国疫苗株无论是核苷酸序列还是氨基酸序列的同源性均最高,而与中国街毒(CDX-1)的核苷酸序列同源性最低,与CVS株的氨基酸序列的同源性最低。Two fragments representing the main regions of glycoprotein (G) gene of deer rabies virus (strain 8202) were amplified by reverse transcriptase polymerase chain reaction(RT PCR) and cloned at Sma I site of pUC18 by blunt end ligation. The two fragments, which were responsible for the virulence of virus and the combination of virus with the receptor of nerve cell, as well as the induction of neutralization antibodies in host organism, are about 800 bp nucleotide(range from 370 to 1 179) in ectomembrane segment of G gene. The sequence of the cDNA was obtained by Sanger′s sequencing technique with their amino acid sequence being deduced. Comparison of the cDNA sequence and deduced amino acid sequence of strain 8202 with those of other 10 strains previously published in the world were performed by computer with DNASIS software. The results showed that the nucleotide homology of them ranged from 73.0% to 96.4% and the deduced amino acid sequence homology, from 86.4% to 94.4%. Both cDNA sequence and deduced amino acid sequence homology of strain 8202 with Chinese vaccine strain(strain 3aG) are the highest; while the nucleotide sequence homology of strain 8202 with Chinese street strain (CDX 1) is the lowest and the deduced amino acid sequence homology of strain 8202 with strain CVS is also the lowest. The results are important for the study on suitable genetic engineering vaccine against deer rabies and the investigation of molecular epidemiology of rabies virus.
