摘  要:目的:探讨食管癌高发区人胎儿发育过程中食管上皮组织抑癌基因p21和增殖细细胞核抗原(proliferation cell nuclear antigen,PCNA)的蛋白表达特征及其与细胞增生的关系。方法:采用免疫组化ABC法检测26例3-10个月胎龄的胎儿食管上皮组织中p21和PCNA的表达状况。结果:在26例食管癌高发区人胎儿食管上皮组织中,p21蛋白总阳性率为42%(11/26),p21蛋白阳性率在低月龄组中明显低于高月龄组,P〈0.05。PCNA单位面积平均阳性细胞数为(450±156)/mm^2,PCNA单位面积阳性细胞数在低月龄组中明显高于高月龄组,P〈0.05。结论:食管癌高发区人胎儿食管上皮组织中p21基因的表达和PCNA单位面积阳性细胞数与胎龄密切相关,可以客观反映食管癌高发区人胎儿食管上皮的组织分化状态。OBJECTIVE: To characterize the expressions of PCNA and p21 in the development of human embryogenic esophageal epithelium and their relationship with cell proliferation from the subjects in high incidence areas for esophageal cancer. METHODS: Immunohistochemical avdin-biotin peroxidase complex (ABC) metheds were used to detect the expressions of PCNA and tumor suppressor gene p21 in embryogenic esophagus epithelium. RESULTS: Of the 26 embryogenic esophageal epithelial specimen from the high incidence area for esophageal cancer, the positive immunostainity rate for p21 was 42%(11/26). There were significant differences (P〈0. 05) in p21 positive rate between early months embryogenic of esophageal epithelium and late stage months. The similar results were observed for PCNA immunostainity in the early and late months of embryogenic esophageal epithelium, P〈0.05. The positive immunostainity cell number for PCNA was (450±156)/mm^2. The positive immunostainity cell number in early months was higher than that in the late months, P〈0.05. CONCLUSION: The expressions of PCNA and p21 may reflect the active proliferative status of embryogenic esophageal epithelium in high incidence areas for esophageal cancer.
