霍奇金淋巴瘤R-S细胞PTEN基因的表达与意义

摘　　要：目的:研究PTEN基因在霍奇金淋巴瘤(Hodgkin's lymphoma,HL)R-S细胞中的表达和意义。方法:PCR检测HL R-S细胞PTEN基因的外显子以证明PTEN基因是否存在,用原位杂交和免疫组化染色方法检测HL R-S细胞PTEN的表达。结果:20例HL中,PCR均检测到PTEN基因的3、4、5外显子,75.0%(15/20)HL R-S细胞中表达PTEN mRNA较强,同样地,70.0%(14/20)HL R-S细胞中PTEN蛋白表达强,而周围小的淋巴细胞不表达或弱表达PTEN mRNA和PTEN蛋白。结论:PTEN的强表达与R-S细胞形成有密切的关系。OBJECTIVE.. To study the expression and significance of PTEN gene in R-S cells of Hodgkin＇ s lymphoma. METHODS： PCR was done to prove the existence of PTEN gene in Hodgkin＇s lymphoma. The expression of PTEN mRNA in R-S cells was detected by hybridization in situ. The expression of PTEN protein in R-S cells was detected by immunohistochemistry. RESULTS.. Exons-3,4, 5 of PTEN genes were found by PCR in all of 20 Hodgkin＇s lymphoma cases. The strong expression of PTEN mRNA was found in R-S cells of 75%（15/20） Hodgkin＇s lymphoma. Likewise, and the strong expression of PTEN protein was found in R-S cells of 70 % （14/20） Hodgkin＇ s lymphoma. In the contrast, the small lymph cells around the R-S cells showed the negative or weak PTEN mRNA and protein expression. CONCUSION： The strong expression of PTEN has close relationship with the formation of R-S cells in Hodgkin＇ s lymphoma.
