摘  要:目的:研究骨肉瘤组织中VEGF和BMP-2的表达及微血管密度(microvessel density,MVD)与转移的关系。方法:用免疫组织化学法检测28例骨肉瘤组织中VEGF、BMP-2的表达和MVD。结果:骨肉瘤组织中VEGF的阳性信号主要位于细胞胞质,BMP阳性信号呈胞质型。VEGF、BMP-2表达和MVD与肿瘤转移密切相关(χ2=15.731,P=0.000;χ2=9.257,P=0.006;t=6.071,P=0.001),VEGF表达与MVD密切有关(t=3.677,P=0.006),VEGF的表达强度与BMP-2表达密切有关(χ2=9.257,P=0.006)。结论:在骨肉瘤的发生发展中VEGF是促进微血管生成的主要细胞因子;VEGF可能影响BMP的骨诱导过程;VEGF和BMP的表达水平与骨肉瘤的转移发生有关,检测VEGF表达BMP-2与MVD值可作为判断骨肿瘤预后的重要指标。OBJECTIVE:To study the expressions of vascular endothelial growth factor and bone morphogenetic protein and microvessel density in osteosarcomas and their correlation with metastasis. METHODS: The immunohistochemistry technique was used to detect the expressions of VEGF and bone morphogenetic protein-2 and MV density in 28 osteosarcoma tissues. RESULTS: In osteosarcoma tissues, the positive signal of VEGF was mainly localized in cytoplasma of tumor cells. The positive signal of bone morphogenetic protein-2 was localized in cytoplasma. VEGF expression, BMP-2 expression and MVD had significant correlation with metastasis Х^2=15. 731, P= 0.000:Х^2=9.257,P=0.006; t=6.071,P=0.001). VEGF expression had significant correlation with MVD (t = 3. 677, P = 0. 006). VEGF expression had significant correlation with BMP-2 expression (Х^2= 9. 257, P = 0. 006). CONLUSlONS: VEGF may be a main factor stimulating angiogenesis in osteosarcoma genesis and development. It is possible that VEGF has an influence on the bone-forming progression of BMP. The expression levels of VEGF and bone morphogenetic protein-2 were related to metastasis of osteosarcoma. Detection of VEGF and BMP-2 expressions and MV density may be useful for judging prognosis of malignant bone tumors.
