小鼠肺腺癌细胞系四株克隆化亚系的转移潜能研究

摘　　要：本文对小鼠肺腺癌细胞系（ＬＡ－７９５）进行单细胞克隆化培养，分离；经筛选获得具有裸小鼠体内移植自发肺转移力相异的４株克隆化亚系，说明母系确实存在转移潜能的异质性。体外实验比较此四株亚系的结果显示：癌细胞的转移潜能不仅与其细胞增殖、分裂及胞浆骨架分布的不均、紊乱程度有关（Ｐ＜０．０１），而且与其胞浆骨架聚集于胞核一侧及内源性非特异性酯酶的活性表达有关（Ｐ＜０．０１）：同时，高转移潜能亚系比低转移潜能亚系出现较低的移动性和粘连性（Ｐ＜０．０１）。提示：癌细胞的胞浆骨架分布不均、紊乱、聚集于胞核一侧及内源性非特异性酯酶的高表达，可能是高转移性癌细胞的重要生物学表型；癌细胞的体内转移力不一定与其体外移动性、粘连性呈正相关。Four cloned cell sublines, isolated ,from mouse lung adenocarcinoma cell line (LA - 795) by single cell technique, were transplanted into the webbed toe of nude mice for 30 days to observe metastasis. The results showed that the four sublianes had a lung metastatic potential differently in the nude mice. This result strongly supported that the par- ent cell line (LA - 795) was of the metastatic heterogenicity - preexisted. In these sublines, the metastasis in vivo was not only correlated to proliferation, nitosis or the irregular, uneven. disorder distribution of cytoskeleton, but also correlated to the charactertistic of the tytoskeleton gathering to one side of the cell nucleu and the endogenous non - specific esterase expression But the metastasis was negatively correlated to the motility of adhesion. Conclu- sion: The irregular, uneven, disorder and gathering to one side or cytoskeleton distribution and highe, expression of the endogeaous non - specific esterase in tumor cell were the important metastasis phenotype of malignant tumor cell possible, but the tumor cell metastatic potential in vivo might be not positively correlated to the tumor cell motility or adhsion in vitro
