摘  要:目的研究中药露蜂房纯化蛋白(nidus vespae protein-Ⅱ,NVP-Ⅱ)对人早幼粒白血病HL-60细胞的生长抑制作用。方法采用NVP-Ⅱ在体外作用于HL-60细胞,经光镜、电镜、荧光显微镜观察形态学变化,TdT缺口末端标记(TUNEL)技术检测NVP-Ⅱ对HL-60细胞增殖、凋亡的影响。结果①光镜下观察到,与生理盐水对照组比较,NVP-Ⅱ处理组HL-60细胞数量减少,胞质内颗粒增多。②透射电镜下发现,NVP-Ⅱ处理组HL-60细胞核染色质靠核膜边集,呈块状或半月形;有的细胞核固缩、断裂为质膜包绕的碎块,有凋亡小体出现。③荧光显微镜下观察到,NVP-Ⅱ处理组HL-60细胞核染色质高度凝聚,致密浓染,或裂解呈碎块状,边缘光滑清晰,呈现典型的凋亡形态特征。④TUNEL检测见阳性细胞核深染,NVP-Ⅱ处理组与生理盐水处理组比较,凋亡细胞明显增多(P<0.05)。结论NVP-Ⅱ可抑制HL-60细胞增殖并诱导其凋亡。Objective To study the inhibition effect of purified protein extracted from Nidus Vespae(NVP- Ⅱ )on human leukemia HL-60 cells. Methods HL-60 cells were treated with NVP-Ⅱ in vitro. Light microscope, electron microscope, fluorescence microscope and TUNEL were used to investigate the proliferation and apoptosis. Results (1)Compared with saline control group, the number of HL-60 cells in NVP-Ⅱ group decreased and granules increased in cytoplasm after being cultured for 72 hours. QNuclear chromatin condensed and marginized to be nubby and formed apoptotic body in HL-60 cells treated with NVP-Ⅱ under transmission electronic microscope. (3) Morphological changes of HL-60 cells in NVP-Ⅱ group such as compaction, condensation and segregation of the nuclear chromatin were observed by fluorescence microscope. (4) Positive cells of hyperchromation were seen with TUNEL. Apoptosis cells increased significantly in NVP-Ⅱ group comparing with saline control group( P 〈 0.05 ) . Conclusion NVP-Ⅱ can inhibit the proliferation and induce the apoptosis of HL-60 cells line in vitro.
