摘  要:目的:研究千斤藤碱对喉癌及鼻咽癌的诱导分化作用。方法:在体外细胞培养的基础上,观察细胞形态学、细胞增殖动力学的变化。采用ABC免疫组化结合图像分析仪及流式细胞仪的方法,定量检测了癌基因C-myc、Bcl-2及抑癌基因p53蛋白。结果:经无毒剂量的千斤藤碱及维甲酸处理后,喉癌及鼻咽癌细胞株的细胞形态趋向良性分化,细胞接触抑制部分恢复,集落形成率和细胞增殖指数明显降低,癌基因C-myc、Bcl-2蛋白明显降低,p53蛋白明显增高。结论:千斤藤碱及维甲酸对喉癌及鼻咽癌细胞株有诱导分化作用,其机理可能是通过抑制癌基因的表达,增加抑癌基因的表达,改变细胞的形态结构和生物学特性,促进癌细胞的分化。Objective: To investigate stepholidine (ST) and retinoic acid (RA) induced differentiation of the cell lines of laryngeal and nasopharyngeal carcinoma. Methods: After the cultured carcinoma cell lines were treated with ST or RA for 5 days, the morphologic changes of the cells were observed with optical and transmission electron microscopy. In addition, the expression of oncogene and carcinoma suppresser gene was determined with ABC immunohistochemistry, flow cytometry and image analysis. Results: After the treatment of ST or RA, the carcinoma cells became well differentiated, their contact inhibition was partially recovered and the colonyforming rate significantly decreased. The contents of the oncogenes C myc and Bcl 2 were remarkably decreased whereas that of the tumor suppresser gene p53 increased. Conclusion: ST and RA can induce the differentiation of the cell lines of laryngeal and nasophryngeal carcinoma through inhibiting the expression of oncogenes and improving the expression of carcinoma suppresser gene.
