摘  要:目的:观察大鼠脊髓损伤(SCI)后的超微病理变化及垂体腺苷酸环化酶激活肽(PACAP)的分布。方法:建立NYU大鼠SCI模型,于术后1 d取损伤节段及其上、下节段脊髓组织,包埋后作超薄切片,胶体金标记及常规铅铀染色后透射电镜观察。结果:电镜下见神经元胞浆及部分神经纤维中有胶体金颗粒,线粒体、内质网等细胞器水肿变性,神经细胞细胞膜、核膜不整齐,髓鞘扭曲变形,板层松解。结论:SCI后PACAP的表达可能与机体自身修复有关。Objective: To observe the ultrastructural pathological changes and the localization of pituitary adenylate cyclase-activating polypepfide(PACAP)in rats after spinal cord injury(SCI). Methods: SCI models of rats were prepared, the spinal cord materials were gotten one day after SCI, the ultrathin sections were made after embedding, the colloidal gold labeling method and lead-uranium staining were used, the ultrastructure of spinal tissue was observed by transmission electron microscope. Results: The colloidal gold granules were found in the neuronal kytoplasm and many nerve fibers. After SCI, the mitochondria swelling and vacuolar degeneration, membrane in bad order, myelin sheath deformation and layers segregation were viewed. Conclusion: As a neurotrophic factor, PACAP may have some relations with self-recovery after SCI.
