摘  要:目的:研究体外培养大鼠视皮层神经元GABA激活电流的特点。方法:用膜片钳记录仪采用全细胞模式记录体外培养大鼠视皮层神经元GABA激活电流。结果:大部分受检细胞可记录到明显去敏感的内向电流,且具有剂量依赖性。IGABA可被足量bicuculline完全阻断。GABAA受体的失敏恢复时间为5min。结论:体外培养视皮层神经元GABA激活电流的特点为探讨视觉形成及其发育机制提供了电生理学依据。AIM: To study the electrophysiological characteristics of gamma-aminobutyric acid (GABA)-activated currents in vitro cultured visual cortex neurons from rats. METHODS: The patch clamp was used to record the whole cell currents of GABA-A channels in vitro cultured visual cortex neurons of 11-15d from the neonatal rats. RESULTS: It was found that majority of the examined neurons were sensitive to GABA. GABA could induce an inward current which was dose dependent. GABA-activated currents could be blocked completely by sufficient bicuculline. The recovery time of GABA-A receptors from desensitization was 5 minutes. CONCLUSION: The study of characteristics of GABA- activated currents provides electrophysiological evidence for the formation and developmental mechanism of vision.
