摘 要:目的研究SSTR2、SSTR5在胃癌中的表达及其与临床病理因素之间的关系,探索SSTR2、SSTR5在胃癌发生、发展以及抑癌机制中的作用。方法对40例手术及胃镜活检取得的胃癌及非癌组织标本、29例手术后癌旁组织标本应用免疫组织化学方法,20例新鲜胃癌、癌旁及非癌组织标本应用Western blot法,检测组织中SSTR2、SSTR5的表达。结果①胃癌组的SSTR2、SSTR5阳性表达较非癌组、癌旁组明显减少,差异有统计学意义(P<0.05);非癌组SSTR2、SSTR5的阳性表达与癌旁组相近,差异无统计学意义(P>0.05);Western blot法分析也得出相同结果。②胃癌标本中SSTR2、SSTR5的阳性表达率分别为32.5%和15.0%,SSTR2、SSTR5的表达与淋巴结转移、分化程度有关(P<0.05),而与其他临床病理因素无关。结论SSTR2、SSTR5的减少可能在非癌组织向胃癌转化的过程中发挥一定作用。SSTR2、SSTR5的表达减少与胃癌有无淋巴结转移、分化程度有关,可作为判断胃癌预后的指标。Objective To investigate the expression of SSTR2 and SSTR5 in gastric cancer and their clinical significance, and to explore the role of SSTR in the process of gastric carcinogenesis and inhibition of tumor growth. Methods 40 specimens of gastric cancer tissue with some adjacent non - cancerous tissue or normal mucosa taken from 29 surgical and 11 biopsy cases, were studied by immmunohistochemistry (SP) and Western blot to determine the expressions of SSTR2 and SSTR5 in them. Results (1) The expressions of SSTR2 and SSTR5 in gastric cancer were lower than that in the adjacent tissue and nontumorous tissue ( P 〈 0.05). The expressions of SSTR2 and SSTR5 in the adjacent tissue were slightly lower than that in the normal mucosa, but the differences were insignificant ( P 〉 0.05 ). These results were supported by the D values of Western blot. (2)The lower expressions of SSTR2 (32.5%) and SSTR5 ( 15.0% ) were found to be significantly correlated with lymph - node metastasis and low differentiation of cancer tissue ( P 〈 0.05 ). Conclusions The expressions of SSTR2 and SSTR5 in gastric cancer tissue are significantly lower than that in its adjacent tissue and the corresponding superficial non - tumorous tissue. The results suggest that SSTR2 and SSTR5 may intervene in the process of gastric carcinogenesis via their negative regulatory effects. The expression of SSTR2 and SSTR5 may serve as a prognostic factor of gastric carcinoma.
