摘　　要：目的探讨血管内皮细胞生长因子(VEGF)在结肠癌中的表达及意义。方法应用实时荧光定量PCR和免疫组织化学技术分别检测结肠癌肿瘤组织和正常人结肠组织中VEGFmRNA及其蛋白水平的表达情况,用CD34单克隆抗体标记肿瘤血管计算其微血管密度(MVD),用SPSS10.0软件分析VEGF表达与该肿瘤微血管密度、临床特征及预后的关系。结果(1)40例结肠癌肿瘤组织中有32例(80.0%)VEGF mRNA表达上调,28例(70.0%)肿瘤细胞VEGF蛋白表达阳性;(2)VEGF表达与该肿瘤微血管密度呈正相关(r=0.7,p=0.007);(3)VEGF表达与该肿瘤的淋巴结转移、组织学分型、Duke s分期有关(p=0.011,p=0.006和p=0.001);(4)生存分析显示,VEGF表达上调为预后不良因素。结论VEGF在结肠癌肿瘤组织中广泛表达,可能与该肿瘤的临床特征和预后有关。Objective To investigate the expression of vascular endothelial growth factor （VEGF） and its clinical significance in human colorectal carcinoma. Methods VEGF mRNA and expression of VEGF protein were detected by real-time fluorescence quantitative PCR method and immunohistochemistry, respectively. Microvessel density （MVD） was counted by detection microvessel with CD34 antibody. SPSS10.0 statistical software was used to analyze the correlations between the expression of VEGF and MVD, clinical features and prognosis of human colorectal carcinoma. Results （ 1 ） Over expression of VEGF mRNA was found in 32/40 （80.0%） by real-time fluorescence quantitative PCR method and the tumor cells expressed VEGF protein in 28/40 （70.0%） by immunohistochemistry; （2） The expression of VEGF was positively correlated to the MVD of the tumor（ r = 0.7 ,p = 0. 007） ; （3） VEGF expression was correlated to lymph nodes metastasis, tissue type and Duke＇s stage （p = 0. 011, p = 0. 006, p = 0. 001 ） ; （4） Positive VEGF expression was independent prognostic factors. Conclusion VEGF was widely expressed in human colorectal carcinoma tissues and it may be related to clinical features and prognosis of this tumor.
